Herein, a rapid and highly efficient method for the synthesis of a new series of pyrimidine derivatives was demonstrated. The strategy was emanated from the reaction of hydrazinyl pyrimidine derivative (1) with different electrophilic species such as ethyl acetoacetate, ethyl 4,4,4‐trifluoro acetoacetate, and phenyl isothiocyanate following cyclocondensation mechanism to afford the corresponding derivatives (2‐6). Furthermore, condensation of hydrazine derivative (1) with different carbonyl compounds via conventional heating and microwave irradiation conditions was employed as a source of Schiff base derivatives bearing pyrimidine moiety (7‐12). The structural features of all newly synthesized compounds were characterized by elemental and spectroscopic evidences. Some of the synthesized compounds were evaluated for in vitro cytotoxicity. The preliminary screening results showed that most of the tested compounds have moderate cytotoxic activity against HepG2 and HCT‐116 cell lines. Finally, a molecular docking study was conducted to reveal the probable interaction with the thymidylate synthase enzyme.

A novel tripodal Schiff base ligand, 5‐amino‐2,4,6‐tris(4‐carboxybenzimino)‐1,3‐pyrimidine (TTPS), was synthesized for the first time by the reaction of 2,4,5,6‐tetraaminopyrimidine with 4‐carboxybenzaldehyde. The ligand was characterized by means of ultraviolet–visible, Fourier transform infrared, one‐dimensional 1H‐NMR, and one‐dimensional 13C‐NMR spectroscopies and elemental microanalysis. The Fourier transform infrared and NMR studies revealed that one of the NH2 group in 2,4,5,6‐tetraaminopyrimidine is not involved in bonding. The in vitro antimicrobial activities of the ligand were investigated against Gram‐negative bacteria: Escherichia coli (ATCC 6749) and Pseudomonas aeruginosa (ATCC 9027), Gram‐positive bacteria: Staphylococcus aureus (ATCC 6538P) and Bacillus cereus (ATCC 14579), and fungi: Candida albicans and Aspergillus niger by the agar well diffusion technique. TTPS exhibits good activity against the test microorganisms. The minimum inhibitory concentrations of TTPS against S. aureus were compared with tetracycline and gentamicin, which are conventional bacterial drugs. The upper level of Lorke's method was used to determine the acute toxicity of the compound. The acute toxicity test indicates that TTPS is toxic at doses above 2154 mg/kg. The in vivo antimalarial assay was carried out on Plasmodium berghei based on the 4‐day suppressive method. The result shows a general dose‐dependent significant parasitemia inhibition compared with the negative control with TTPS having inhibition of 72.20% at 50 mg/kg and 65.81% at 25 mg/kg close to the value (87.22%) of the standard drug artesunate 5 mg/kg. Density functional theory method was used to complement this experimental investigation.

A series of novel binuclear transition metal complexes was synthesized by reaction of a Schiff base ligand 2-((1-methyl-2-((2-oxo-1,2-dihydro-pyrimidin-4-yl)imino)propylidene)amino)acetic acid) (L a H) derived from 4-amino-1H-pyrimidin-2-one, diacetyl, glycine and the corresponding chloride salt of Cu(II), Ni(II), Co(II) and Zn(II) metals in a 1:1 (metal:ligand) mole ratio. The compounds were characterized by elemental analyses, molar conductance measurements, magnetic moment measurements and various spectral studies viz. IR, UV-Vis, 1 H-NMR, 13 C-NMR, EPR and ESI-MS. The molar conductance measurements revealed the non-electrolytic nature of the metal complexes. Electronic absorption spectral data, electronic paramagnetic resonance parameters and magnetic moment values revealed an octahedral geometry for the binuclear metal complexes. A cyclic voltammetric study of Ni(II) complex shows a couple of one-electron anodic responses near 0.70 and 1.10 V. The in vitro biological activity of the Schiff base ligand and the binuclear complexes was assessed against bacteria (Staphylococcus aureus, Bacillus subtilis, Escherichia coli and Salmonella typhi) and fungi (Candida albicans and Candida parapsilosis) to ascertain their antibacterial and antifungal properties.
