“…In some cases, Ru II polypyridyl complexes were found to photocleave DNA by sensitization of singlet oxygen ( 1 O 2 ) [15]. In others, 1 O 2 and superoxide anion radical (O .À 2 ) may play an important role in the photocleavage [9] [10] [30]. On the other hand, Ru complexes as antitumor drugs have attracted great attention; complex [H 2 im] [trans-RuCl 4 (dmso-S)(Him)] (NAMI-A; Him ¼ 1H-imidazole, dmso ¼ dimethyl sulfoxide) has entered clinical trials due to its very good antimetastatic activity [31] [32].…”

“…[ [30]. When circular plasmid DNA is subjected to electrophoresis, relatively fast migration will be observed for the intact supercoil form (Form I); if scission occurs on one strand (nicking), the supercoil will relax to generate a slower-moving open circular form (Form II); if both strands are cleaved, a linear form (Form III) that migrates between Form I and Form II will be generated [48].…”

“…2) showed a strong decrease in intensity (hypochromicity: 20.6% for complex 1 and 28.5% for complex 2) and a small red shift (3 nm for complex 1 and 2, respectively). The change in the absorbance of the MLCT band with increasing amounts of CT-DNA was used to derive the intrinsic [37], but smaller than the classical intercalators whose binding constants were in the order of 10 7 [38]. The different DNA-binding properties of complexes 1 and 2 are due to the different ancillary ligand.…”

“…Therefore, development of GC-targeting probes is essential for the locating and reparation of DNA damage. To design novel, sensitive DNA molecular luminescent probes for GC sequences, we have prepared a guanine-functionalized, dppz-based Ru II -complex [Ru(bpy) 2 2 þ with calf-thymus (CT) DNA, poly(dA) · poly(dT), and poly(dG) · poly(dC) was studied [73]. Viscosity measurements and DNA thermal denaturation experiments suggest that, upon modification of dppz ligand, [Ru(bpy) 2 (appo)] 2 þ preserves the high DNA-binding affinity of its parent complex [Ru(bpy) 2 (dppz)] 2 þ , and binds CT-DNA, poly(dA) · poly(dT), and poly(dG) · poly-(dC) in an intercalative mode.…”
