Synthesis, self-assembly, and immunological activity of α-galactose-functionalized dendron–lipid amphiphiles

Abstract: Nanoassemblies presenting multivalent displays of biologically active carbohydrates are of significant interest for a wide array of biomedical applications ranging from drug delivery to immunotherapy. In this study, glycodendron-lipid hybrids were developed as a new and tunable class of dendritic amphiphiles. A modular synthesis was used to prepare dendron-lipid hybrids comprising distearylglycerol and 0 through 4th generation polyester dendrons with peripheral protected amines. Following deprotection of the a… Show more

“…Self-assembling of arborescent neoglycodendrimersomes harboring α- C -linked galactopyranosides mounted on a 2,2-bis(hydroxymethyl)-propionic acid (bis-HMPA) scaffold ending with palmitic acid dimers has been described by the Gillies group [ 37 ] ( Scheme 7 ). The glycolipid hybrid was made to mimic the natural KRN 7000 immunostimulant ( Figure 2 ).…”

“…Amphiphilic dumbbells are constituted of two flexible dendrons coupled on each side of a rigid hydrophobic core [34,35,36,37,38,39,40]. Amphiphiles with a non-dendritic hydrophobic structure (for instance, one alkyl chain (Figure 1d) or p -phenylene chains (Figure 1e)) connected to a hydrophilic dendron can be classified as dendritic amphiphiles [32,33,41,42,43,44,45,46,47].…”
