Twenty three CPTs (Candidate Plus Trees) of Pongamia pinnata were selected from different agro-climatic conditions of Haryana state of India and were assessed to identify the elite planting material for improvement of the species in terms of oil content. The differences among CPTs of P. pinnata were significant for seed oil content and all growth parameters of the progenies of these CPTs at the seedling stage. Oil content in P. pinnata varied from 27.07 (P12) to 38.17% (P2). The estimates of genotypic coefficients of variation for the characters studied were less as compared to the phenotypic coefficients of variation for all the characters examined. The highest phenotypic coefficient of variation (49.33) and genotypic coefficient of variation (28.56) was recorded for the germination percentage followed by height of the first branch. Number of leaves (0.5551**), inter-nodal length (0.5580**) and number of branches (0.6182**) showed high and positive correlation with the seed oil content. The progeny number 9, 21 and 2 were found to be the best on basis of oil content (36. 37, 36.83 and 38.17 %, respectively), and other characters examined. D 2 analysis grouped the CPTs into 5 clusters. The highest numbers of progenies were included in the cluster I followed by cluster III and least number of progenies i.e., two were observed in cluster II. The intra cluster distances ranged from 4.12 (cluster V) to 5.96 (cluster II). The maximum inter-cluster distance was observed between cluster II and III (10.02) followed by I and III and minimum was between clusters I and cluster V. The crosses between clusters II and III may result sufficient segregation for further improvement of the species. Therefore, the progenies belonging to the clusters II and III could be taken as parents for a successful hybridization program.

Pongamia pinnata, commercially important tree species used to produce biofuels, is known for its multipurpose benefits and its role in agro-forestry. Present study examines the amenability of vegetative propagation and effect of maturation in candidate plus tree P. pinnata through rooting of stem cuttings treated with varying concentrations and combinations of auxins. The performance of the cuttings was evaluated using SAS GLM software and the data were analyzed as a one-way classified data with and without sub sampling for inferring auxin concentration that can be included in programmes aimed at genetic improvement of the tree species. All auxin treatments promoted sprouting and at lower concentrations triggered/enhanced rooting of cuttings. The effectiveness was in the order of IBA [ NAA [ IAA when applied singly. IBA at 4.92 mM was found to be most effective where rooting percentage and number of roots were significantly higher (P \ 0.01) than in control. However higher concentrations of auxins above 7 mM in general inhibited the rooting of cuttings. The interaction among auxins was found to be effective in root induction and differentiation and the most stimulating effects were observed in three-component mixture. The effect of other cutting characteristics such as juvenility and cutting position on rooting is also discussed.

Pongamia pinnata (Pongamia) is a multipurpose tree that has been used traditionally for medicine, green manure, insecticide, fuel and also a source of timber. It is an indigenous plant to the Indian subcontinent and Southeast Asia, and has been successfully introduced to Northern Australia, New Zealand and several other countries with humid tropical lowlands. Recently, Pongamia has attracted interest as one of the alternative feedstock for biodiesel productions due to its ability to produce seeds with high oil content. The fatty acid methyl ester (FAME) composition of Pongamia met the biodiesel specification standards of USA (ASTMD 6751), Germany (DIN 51606) and European Standard Organisation (EN 14214), and is considered as one of the most suitable sources of biodiesel. Despite the positive attributes described above, Pongamia is not a domesticated species and further research is required for the selection of elite cultivars with desirable traits optimal for large scale production of Pongamia oil. In addition, limited information is currently available regarding the flowering mechanism of Pongamia at the molecular level that could provide the platform for the development of oil-rich seeds.The aim of this thesis is to provide insight and increase the understanding of the genetic network that leads to flowering in Pongamia. Based on large phenotypic diversity of Pongamia, this information could subsequently be used to determine and select traits with unique flowering time that will allow a harvesting window, where harvesting process will not clash with flowering. The first objective of this project is to identify, isolate and characterise the candidate genes that regulate flowering time in Pongamia. This was achieved using molecular biology techniques and bioinformatic approaches based on the information available on flowering mechanisms of Arabidopsis and soybean in the literature. The second objective of this project is to determine the expression pattern of isolated genes during flowering onset. This was achieved through transcriptome methodologies such as RT-PCR and qRT-PCR. The third objective of this project is to investigate the genetic variety of Pongamia. This was achieved through Single Nucleotide Polymorphism (SNPs) analysis and its correlation with the late flowering phenotype of Pongamia.PpCOL-1 and PpPHYB are the two genes that were investigated in this thesis. The orthologue genes of PpCOL-1 and PpPHYB are involved in the flowering regulation of the annual plant Arabidopsis.Since Arabidopsis and Pongamia are both long day plants, it is postulated that these genes might play a role in the flowering time control in Pongamia. PpCOL-1 and PpPHYB were successfully isolated, characterised and compared to their respective orthologue genes in Arabidopsis, Soybean and Medicago.
