Systemic lidocaine inhibits high-mobility group box 1 messenger ribonucleic acid expression and protein in BALB/c mice after closed fracture musculoskeletal injury

Abstract: Background:Severe musculoskeletal trauma can trigger an inflammatory response, and an excessive inflammatory response can lead to systemic inflammatory response syndrome and multiorgan failure. High-mobility group box 1 (HMGB1) is an early mediator pro-inflammatory cytokine in sterile injuries and a late cytokine mediator in infection and sepsis. Previous research has shown that administration of systemic lidocaine can inhibit HMGB1 expression in macrophages of septic rats. The aim of this study was to demonst… Show more

“…The samples that met the requirements were brought to the Laboratory of Molecular Biology and Immunology Faculty of Medicine Universitas Hasanuddin in Makassar for the examination of the mRNA gene Treg, Treg protein levels, CD4 + levels, and serum CD8 + levels. The RNA extraction used the Guanidium thiocyanate and diatom method [ 10 , 11 ].…”

“…The groin of the mice was clamped firmly with one needle holder for fixation, and the middle of the femur was also clamped with the other needle holders. The left thighbone of the mouse was broken by moving the needle holders located in the middle of the thigh up and down against the needle holders in the groin until a cracking sound was heard and the fracture of the bone in the thigh when moved by hand was felt [ 29 ]. It was stabilized with external fixation and the mice were then returned to their cages.…”
