T-cell epitope vaccine design by immunoinformatics

Abstract: Vaccination is generally considered to be the most effective method of preventing infectious diseases. All vaccinations work by presenting a foreign antigen to the immune system in order to evoke an immune response. The active agent of a vaccine may be intact but inactivated (‘attenuated’) forms of the causative pathogens (bacteria or viruses), or purified components of the pathogen that have been found to be highly immunogenic. The increased understanding of antigen recognition at molecular level has resulted… Show more

“…CD4 + and CD8 + T-cells are activated by sensing short peptides presented within MHC class II and class I context respectively. Researchers now harness Immunoinformatics tools to predict potential CD4 + /CD8 + T-cell epitopes in silico and then select in vitro/in vivo evaluated immunogenic ones to arrange them together in a polytope construct [10,11]. Polytopes or T-cell vaccines as novel subunit vaccines are highly preferred since they carry minimal immunogenic part of the protein where whole protein might have gift by Prof. Genevieve Milon, Pasteur Institute of Paris) were cultured for weeks to concentrate secreted antibody in about 1500 ml medium.…”

“…Analysis of enhanced RV data through decision trees provided a list of 233 non-redundant set of most probable vaccine candidates from 23 strains and species of Mycobacteria. Recent trends of vaccinologists aim for epitope based vaccines (Patronov and Doytchinova 2013;Khan et al 2007). Towards facilitating these efforts, we analyzed the information on epitopes in terms of their conservation among orthologs (Khan et al 2007).…”

“…On the other hand, EHEC immunity requires humoral and T helper cell responses. Generally, analysis of the binding affinity of antigenic peptides to MHC molecules is the essential object when predicting immunogenic epitopes (Patronov and Doytchinova 2010;Betts and Russell 2003). Identification of epitopes with high binding affinity for MHC class II was performed using IEDB server.…”
