T-cell prolymphocytic leukemia with an unusual phenotype CD4+ CD8+

Abstract: A patient with T-cell prolymphocytic leukemia (T-PLL) is described. The outcome was poor, with death 8 months after diagnosis, despite several therapeutic interventions. The cells carried both CD4 and CD8 epitopes, but other thymocytic markers were absent. The spleen showed infiltration of CD4+ CD8+ prolymphocytes in the red pulp and in T-cell-dependent areas of the white pulp. Immunologic function studies revealed proliferation after stimulation with mitogens and even several antigens. However, in the mixed l… Show more

“…Both cases of ALL were immunophenotyped in suspension and on cytocentrifuge preparations (6) . Primary antibodies used were : goat or burro FITC and TRITC-conjugated anti-Ig antisera (Kallestad, Nordic), for detection of cell surface Ig and cytoplasmic Fr chains, rabbit anti-terminal transferase (TdT) (Bethesda Research Laboratories, Gaithersburg, MD), CD10 (BA3) (Hybritech, Luik, Belgium), CD19 (B4), and CD20 (Bl), CD24 (BAI) (Hybritech), CD2 (Leu-5) (Becton Dickinson & Co.), and CD7 (WT1) (Sanbio, Uden, The Netherlands) .…”

“…Mononuclear cells were obtained from heparinized venous blood and bone marrow by Ficoll-Hypaque gradient centrifugation. The peripheral blood contamination in the aspirates, calculated according to Holdrinet et a1 (1980) phenotypes were determined by immunoluorescense microscopy as described (Kluin-Nelemans et al, 1987). At least 200 nucleated cells were scored.…”
