Tacrolimus Loaded Cationic Liposomes for Dry Eye Treatment

Abstract: Eye drops are ophthalmic formulations routinely used to treat dry eye. However, the low ocular bioavailability is an obvious drawback of eye drops owing to short ocular retention time and weak permeability of the cornea. Herein, to improve the ocular bioavailability of eye drops, a cationic liposome eye drop was constructed and used to treat dry eye. Tacrolimus liposomes exhibit a diameter of around 300 nm and a surface charge of +30 mV. Cationic liposomes could interact with the anionic ocular surface, extend… Show more

“…It has been proven that cationic liposomes are the most promising siRNA carriers with high load rates, biocompatibility, and biodegradability. Commonly used cationic lipids include DOTAP [ 94 ], trimethyl-2 chloride, 3-dioleoxypropyl ammonium (DOTMA) [ 95 ], 3β-[N- (N′, N′-dimethylaminoethyl) aminoformyl] CHO (DC-CHOl) [ 96 ], etc. However, the stability, membrane fusion, and transfection efficiency of liposomes alone are poor, so auxiliary lipids are often necessary.…”

“…Tacrolimus is another widely used immunosuppressant for DED treatment. Biodegradable formulations of tacrolimus, such as cationic liposome [ 113 ] and thermoresponsive in situ gel formulations [ 114 ] have been developed to improve ocular retention, enhance corneal tacrolimus concentration, and decrease levels of reactive oxygen species and inflammatory factors associated with DED ( Table 1 ).…”

“…Liposomes are one of the most popular nanoparticulate drug delivery systems, which are characterized by nanosized artificial vesicles with single or multiple bilayer membranes and an aqueous core, formed by phospholipids and cholesterol [68,69]. Their limited immunogenicity and high biodegradability profiles are attributed to the phospholipids used, which are similar to lipids found in cell membranes [70,71]. Additionally, they show the capacity to load a wide range of therapeutic compounds of different physicochemical properties, from lipophilic compounds loaded in the lipid bilayers to hydrophilic drugs encapsulated in the aqueous core, and also ionic and amphiphilic molecules that can be loaded using cationic or anionic lipids [72,73].…”
