Target deconvolution strategies in drug discovery

Abstract: Recognition of some of the limitations of target-based drug discovery has recently led to the renaissance of a more holistic approach in which complex biological systems are investigated for phenotypic changes upon exposure to small molecules. The subsequent identification of the molecular targets that underlie an observed phenotypic response--termed target deconvolution--is an important aspect of current drug discovery, as knowledge of the molecular targets will greatly aid drug development. Here, the broad p… Show more

“…The former has become increasingly important in view of the recent tendency in the pharmaceutical industry to switch from target-based drug discovery to phenotypic screening where prior knowledge of the target is not a prerequisite. Although various approaches have been developed, target identification remains challenging (40,41). In analogy to the threehybrid assays that have been successfully applied to identify drug targets in yeast (42), we developed a three-hybrid variant of KISS that enables drug-target analysis in living mammalian cells.…”

“…I dentifying molecular pathways targeted by a compound (drug effects), and the specific compound-substrate interactions (drug mode of action-MoA), is of paramount importance for the development of new drugs, and also for new clinical applications of already existing drugs (1)(2)(3). Systems biology approaches are naturally suited to capture the complexity of drug activity in cells (4)(5)(6).…”
