Targeted proteomic approach in prostatic tissue: a panel of potential biomarkers for cancer detection

Abstract: Prostate cancer (PCa) is the sixth highest causes of cancer-related deaths in men. The molecular events underlying its behavior and evolution are not completely understood. Prostate-specific antigen (PSA) is the only approved Food and Drug Administration biomarker. A panel of ten stage-specific tumoral and adjacent non tumoral tissues from patients affected by PCa (Gleason score 6, 3+3; PSA 10 ÷19 ng/ml) was investigated by MS-based proteomics approach. The proposed method was based on identifying the base-sol… Show more

“…Literature data revealed the tri-modality feature for zebrafish ( Danio rerio ) heart proteome, with the majority of proteins centered around pIs 5.5 and 9 and fewer proteins around neutral pI 27 . Furthermore, proteomic data on Gillichthys mirabilis cardiac tissues revealed 37 protein, involved in energy metabolism, mitochondrial regulation, iron homeostasis, cytoprotection against hypoxia, and cytoskeletal organization, with the majority of proteins centered around pIs 5.5–6.5 28 The reliable detection of species-specific proteins and peptides, unique in mass and amino acid sequence, depends on proper protein solubilization, digestion, and sensitive MS analysis 29,30 . Therefore, two different aqueous media were tested for the ability to extract proteins from the goldfish cardiac tissues: phosphate buffer (pH 7.5) and ammonium bicarbonate solution (50 mM, pH 8).…”

“…Many studies on the proteome of PCa, e.g., from Aiello et al [11] , Davalieva et al [12] , or Iglesias-Gato et al [13] , compare malignant to benign tissue. We considered it especially interesting to investigate whether the proteome composition of malignant tissue differs from other malignant tissue.…”

“…MALDI and SDS-Page protein profile of HTP fractions revealed the ubiquitous presence of HSA (Supporting Information, Figs 3S and 4S ), demonstrating the non-specificity of the method. In order to check all fractions by direct mass spectrometry, a novel MS-compatible experimental procedure based on the use of immunoaffinity devices was designed (Experimental session) 18 . MARS cartridge ( III a ) can selectively remove high-abundant proteins from human serum, plasma, and cerebrospinal fluid, offering the opportunity to analyze up to 200 samples with no memory effect.…”
