Targeted Tumor Therapy Based on Nanodiamonds Decorated with Doxorubicin and Folic Acid

Abstract: The fabrication of nanodiamond (ND)-based drug carriers for tumor-targeted drug delivery is described. The ND clusters with an average size of 52.84 nm are fabricated using a simple fluidic device combined with a precipitation method and then conjugated with folic acid (FA) and doxorubicin (Dox) via carbodiimide chemistry to obtain FA/Dox-modified ND (FA/Dox-ND) clusters. Cell culture experiments revealed that KB (folate receptor-positive) cells are preferentially ablated by FA/Dox-ND clusters compared to A549… Show more

“…With increasing ssDNA concentration of the ND-conjugates, zeta potential decreases, which leads to an increased mean agglomerate size due to the reduced repulsion and lower amount of available functional surface groups of the NDs. Recently, the drug delivery of NDs using carbodiimide chemistry [46,47] and electrostatic interactions [36] has been discussed. We show the successful binding of ssDNA to carboxylated NDs based on thioester bridges.…”

“…The engineering of those span from electronics to medicine [1], where the biomedical aspects of nanomaterials are of particular interest [2,3]. Properly designed nanomaterials can serve as scaffolds [4,5], drug [6] or gene [7] carriers, or due to their inherent properties, can be utilized in anticancer therapy [8,9,10]. With constantly growing numbers of cancer incidence [11], tumor treatment has become one of the most investigated topics all around the world.…”

“…A slightly more complex but targeted approach is to add a targeting molecule in addition to the anticancer drug. Ryu et al used folic acid for this purpose . Many cancer cells, including oral cancer (KB) cells that were used in this study, have receptors for folic acid and thus, this molecule helped to achieve targeting.…”
