Targeting Activated Lymphocytes With Photodynamic Therapy: Susceptibility of Mitogen‐stimulated Splenic Lymphocytes to Benzoporphyrin Derivative (Bpd) Photosensitization

Abstract: Benzoporphyrin derivative monoacid ring A (BPD), a hydrophobic chlorin-like porphyrin derivative, which fluoresces strongly at 690 nm, may have potential for both oncologic and nononcologic applications in photodynamic therapy (PDT). To study the influence of cellular characteristics on the uptake of BPD, the murine tumor cell line (P815), and in vitro and in vivo concanavalin A (Con A) -stimulated and unstimulated murine splenic lymphocytes were incubated with 2 micrograms/mL BPD at 37 degrees C for 0-60 min.… Show more

“…Cellular activation affords additional selectivity in therapy to pathogenic tissues; activated cells have been shown to take up BPD-MA to a greater extent than quiescent populations, resulting in greater sensitivity to PDT [14]. We have shown in rabbit synovium, that inflammatory cells bearing the MHC-class II activation marker demonstrated greater uptake of BPD [9].…”

“…Numerous studies have documented that photosensitisers efficiently accumulate in normal human lymphocytes [105,106]. However, resting lymphocytes only poorly accumulate photosensitisers, and the uptake is improved in activated cells [106][107][108]. PDT of resting T-cells leads to MHC class I molecule's downregulation.…”

“…A prototype with up to 1.0 W was employed in the present study. Up to 99% of the BPD-MA is excreted within 72 h; no substantial side effects have yet been described [18]. For treating the macula degeneration of the eye, which is performed as a clinical phase II trial with an identical dose prophylactic, only wearing of dark glasses for 7 days is recommended.…”
