Targeting Dermal Fibroblast Subtypes in Antifibrotic Therapy: Surface Marker as a Cellular Identity or a Functional Entity?

Abstract: Fibroblasts are the chief effector cells in fibrotic diseases and have been discovered to be highly heterogeneous. Recently, fibroblast heterogeneity in human skin has been studied extensively and several surface markers for dermal fibroblast subtypes have been identified, holding promise for future antifibrotic therapies. However, it has yet to be confirmed whether surface markers should be looked upon as merely lineage landmarks or as functional entities of fibroblast subtypes, which may further complicate t… Show more

“…Its responsibilities encompass providing essential structural support to tissues, secreting the extracellular matrix, contributing to the mending of tissue damage, and actively participating in immune responses. [ 22 , 23 ] The intricate process of skin regeneration involves a multifaceted interplay between cells, scaffold structures, and the extracellular matrix, in which fibroblasts play an indispensable role. [ 6 , 7 , 9 , 10 , 24 ] Similar to other organs such as the heart, lungs, gastrointestinal tract, muscles, and adipose tissues, the skin emerges from mesenchymal-derived connective tissues.…”

“…The excessive deposition of ECM proteins (e.g., collagen and fibronectin) by activated fibroblasts and myofibroblasts results in thickening and hardening of tissues (i.e., fibrosis) ( Knuppel et al, 2017 ; Huang et al, 2021 ; Plikus et al, 2021 ). Tissue fibrosis is involved in the genesis of many diseases.…”
