Targeting Nrf-2 is a promising intervention approach for the prevention of ethanol-induced liver disease

Abstract: Alcoholic liver disease (ALD) remains to be a worldwide health problem. It is generally accepted that oxidative stress plays critical roles in the pathogenesis of ALD, and antioxidant therapy represents a logical strategy for the prevention and treatment of ALD. Nuclear factor erythroid-derived 2-like 2 (NFE2L2 or Nrf-2) is essential for the antioxidant responsive element (ARE)-mediated induction of endogenous antioxidant enzymes such as heme oxygenase 1 (HO-1) and glutamate-cysteine ligase [GCL, the rate-limi… Show more

“…Nuclear factor erythroid 2-related factor 2 (Nrf-2) has been shown to up-regulate the expression of the various antioxidants while role of ROS in inflammation in NF-kB mediated inflammation has been studied in cisplatin-induced organ injury [50]. Nuclear factor erythroid 2-related factor 2 (Nrf-2) is a basic leucine zipper transcription factor present in inactive form and associated with cytoskeleton-associated protein, Keap1, in cytoplasm [76]. The activation of this transcription factor is regarded as a crucial molecular target of several chemopreventive agents [77,78].…”

“…The function of Nrf-2 is to prevent oxidative stress via antioxidant response element (ARE)-mediated inductions of numerous phase-2 detoxifying enzymes, specifically HO-1 [78,79]. Generally, the activation of the transcription factor Nrf-2 is induced by cellular oxidative stress and acts as a marker of pro-oxidant stressors and electrophiles [76,80]. Many research groups have shown strong interaction between Nrf-2 and NF-κB [81,82].…”

“…In fact, Nrf-2 is essential for the antioxidant responsive element (ARE)-mediated induction of endogenous antioxidant enzymes such as heme oxygenase 1 (HO-1), NAD(P)H dehydrogenase [quinone] 1 (NQO-1), and GCL. 41 Interestingly, it has been proposed that targeting the heme-heme oxygenase system may prevent severe complications following COVID-19 infection. 42 In parallel, NAC inhibits the oxidative stress-mediated activation of nuclear factor kappa-lightchain-enhancer of activated B cells (NFκB) and biochemical pathways upregulating pro-inflammatory genes.…”

“…In chronic alcohol abuse, alcohol increases the expression of CYP2E1 in liver, causing dysregulations in lipid peroxidation through ROS free radicals. Therefore, it leads to the dynamic imbalance of antioxidant systems which target the important antioxidant regulatory gene nuclear factor erythroid 2-related factor 2 (Nrf-2) in ALD [38]. It also damages other antioxidant genes which have important roles in ROS scavenging, such as superoxide dismutase (SOD), glutathione (GSH), catalase, peroxidase-1, metallothionein, and heme oxygenase (HO-1) [39,40].…”
