Targeting Trichothecene Biosynthetic Genes

Abstract: Biosynthesis of trichothecenes requires the involvement of at least 15 genes, most of which have been targeted for PCR. Qualitative PCRs are used to assign chemotypes to individual isolates, e.g., the capacity to produce type A and/or type B trichothecenes. Many regions in the core cluster (consisting of 12 genes) including intergenic regions have been used as targets for PCR, but the most robust assays are targeted to the tri3 and tri12 genes. Quantitative PCRs, that work across trichothecene-producing member… Show more

“…Conidia were harvested from 5-day-old mungbean liquid medium (MBL) (Wei et al, 2017 ; 30 g mung bean, 1 L ddH 2 O, cook for 10 min, filtered and sterilized) and resuspended at 10 6 conidia/mL in sterile water. For infection on flowering wheat ( cv .…”

“…Trichothecenes are divided into four types (A–D) according to their characteristic functional groups. Types A and B are of greater concern to people due to their high toxicity and frequent presence as food contaminants [ 1 , 2 ]. For both of these reasons, T-2 toxin and deoxynivalenol (DON), which belong to type A and type B, respectively, are considered to be the most important compounds of this class (Figure 1 ) [ 3 5 ].…”

“…Wei et al [79] reported on PCR assays based on the TRI3 gene (Figure 3a,b) and on the TRI12 gene [58,89]. These primers target trichothecene B-producing Fusarium species and the size of the amplicon is indicative of the chemotype of the isolate (Table 3).…”
