Tau and mTOR: The Hotspots for Multifarious Diseases in Alzheimer's Development

Abstract: The hyperphosphorylation of tau protein and the overexpression of mTOR are considered to be the driving force behind Aβ plaques and Neurofibrillay Tangles (NFT's), hallmarks of Alzheimer's disease (AD). It is now evident that miscellaneous diseases such as Diabetes, Autoimmune diseases, Cancer, etc. are correlated with AD. Therefore, we reviewed the literature on the causes of AD and investigated the association of tau and mTOR with other diseases. We have discussed the role of insulin deficiency in diabetes, … Show more

“…S2). This result is in line with others, where mTOR is found to participate in memory formation and consolidation, and seems targeted by Aβ42 (Lin et al, 2014;Mueed et al, 2019;Uddin et al, 2018). Thus, mTOR inhibition may be another experimental strategy, independent from PI3K activation, to be considered in future AD studies.…”

“…Protein misfolding and subsequent aggregation is at the heart of Alzheimer's pathophysiology (Mueed et al, 2019). The mTORC1 complex has been reported to stimulate protein synthesis (Papadopoli et al, 2019).…”

“…Moreover, mTOR mediates tau synthesis and phosphorylation at specific sites and is found to co-localize with NFTs (An et al, 2003;Tang et al, 2013). This results in microtubule instability as tau is an essential microtubule associated protein and its hyperphosphorylation causes it to detach from microtubules (Mueed et al, 2019).…”

“…The mammalian target of rapamycin (mTOR) regulates autophagy. The use of rapamycin or starvation could inhibit mTOR activity and thereby increase autophagy activity [ 104 , 105 ]. In in vitro and rodent AD model studies, mTOR activity was correlated with Aβ accumulation.…”
