ABSTRACT. Phylogenetic analyses of the Hypnales usually show the same picture of poorly resolved trees with a large number of polyphyletic taxa and low support for the few reconstructed clades. One odd clade, however, consisting of three genera that are currently treated either within the Leskeaceae (Miyabea) or Neckeraceae (Homaliadelphus and Bissetia), was retrieved in a previously published phylogeny based on chloroplast rbcL. In order to elucidate the reliability of the observed Homaliadelphus -Miyabea -Bissetia -clade (HMB-clade) and to reveal its phylogenetic relationships a molecular study based on a representative set of hypnalean taxa was performed. Sequence data from all three genomes, namely the ITS1 and 2 (nuclear), the trnS-rps4-trnT-trnL-trnF cluster (plastid), the nad5 intron (mitochondrial), were analyzed. Although the phylogenetic reconstruction of the combined data set was not fully resolved regarding the backbone it clearly indicated the polyphyletic nature of various hypnalean families, such as the Leskeaceae, Hypnaceae, Hylocomiaceae, Neckeraceae, Leptodontaceae and Anomodontaceae with respect to the included taxa. In addition the results favor the inclusion of the Leptodontaceae and Thamnobryaceae in the Neckeraceae. The maximally supported HMB-clade consisting of the three genera Homaliadelphus (2-3 species), Miyabea (3 species) and Bissetia (1 species) is resolved sister to a so far unnamed clade comprising Taxiphyllum aomoriense, Glossadelphus ogatae and Leptopterigynandrum. The well-resolved and supported HMBclade, here formally described as the Miyabeaceae, fam. nov. is additionally supported by morphological characters such as strongly incrassate, porose leaf cells, a relatively weak and diffuse costa and the presence of dwarf males. The latter are absent in the Neckeraceae and the Leskeaceae. It is essentially an East Asian family, with one species occurring in North America.

A number of reviews have accompanied and monitored the progress of molecular phylogenetic research on bryophytes, focusing on the publication record itself, bryophyte phylogeny and systematics in the molecular era, as well as the evolution and phylogenetic utility of markers from different genomes. However, none of the recent reviews include a detailed characterization of all molecular markers used in bryophyte phylogenetics. Here we provide an overview of the history and current state of marker utilization, including coding and non-coding sequence markers from all three genomes as well as fingerprinting approaches. The molecular architecture and evolutionary peculiarities, as well as practical aspects such as amplification and sequencing strategies, are outlined for the DNA sequence markers, with a focus on the most commonly employed regions. Their phylogenetic utility and potential for solving some of the remaining, pressing questions in bryophyte phylogeny, as well as their suitability for molecular species identification by DNA barcoding, are discussed.

The peculiar disjunction between Macaronesia and Australasia of the morphologically isolated pleurocarpous moss genus Echinodium is one of the most prominent questions in bryology. Echinodium as traditionally circumscribed comprises six extant species, four restricted to the Macaronesian archipelagos and two confined to the Australasian/ Pacific regions. Molecular phylogenetic analyses based on plastid trnL UAA intron and nuclear ribosomal ITS sequences indicate that Echinodium is polyphyletic and split into three groups. Three of the four Macaronesian species (E. spinosum and the single island endemics E. renauldii and E. setigerum) are closely related to each other and treated as Echinodium s.str. (Echinodiaceae). Further clarification of the relationships of Echinodium s.str. with Orthostichella, a segregate of Lembophyllaceae, is needed. The remaining Macaronesian species, E. prolixum, is transferred to Isothecium (Lembophyllaceae); this systematic position is also strongly supported by leaf characters. The two Australasian species, E. hispidum and E. umbrosum, are molecularly unrelated to the Macaronesian species and are transferred to Thamnobryum in the Neckeraceae. While the molecular data suggest that the peculiar distribution pattern of 'Echinodium' is an artefact, the striking morphological similarity observed in Macaronesian and Australasian species cannot be dismissed. Possible explanations are: (i) parallel morphological evolution of the 'Echinodium habit' in Macaronesia and Australasia, or (ii) retention of a set of plesiomorphic characters in non-related groups in relict habitats, the Macaronesian laurel forest and the austral temperate rain forests, respectively. Of these hypotheses, the evolutionary parallelism hypothesis seems more plausible for several reasons, which are discussed.
