Temporal evolution of helix hydration in a light-gated ion channel correlates with ion conductance

Abstract: The discovery of channelrhodopsins introduced a new class of lightgated ion channels, which when genetically encoded in host cells resulted in the development of optogenetics. Channelrhodopsin-2 from Chlamydomonas reinhardtii, CrChR2, is the most widely used optogenetic tool in neuroscience. To explore the connection between the gating mechanism and the influx and efflux of water molecules in CrChR2, we have integrated light-induced time-resolved infrared spectroscopy and electrophysiology. Cross-correlation a… Show more

“…The photocurrents decline with a dominant voltage-dependent time constant of 10-22 ms and a second, minor, slow time constant of 70-220 ms ( Fig. 2A and B, top), in general agreement with previous reports (8). Decreasing extracellular Na + not only reduces inward directed photocurrent amplitudes, but also affects the temporal evolution of inward currents ( Fig The proton versus sodium selectivity of all three open states is analyzed at three different time points after the actinic laser pulses (0.1 ms, 5 ms and 100 ms; Fig.…”

“…S3). Interestingly, lightinduced movement of helices B and G has also been found to play an important role in the pore formation of channelrhodopsin (ChR) (28,29). This helix movement correlates with a shift in the electrostatic potential at the opening of the intracellular cavity toward more positive values relative to the K/L state (Fig.…”

“…Retinal isomerization appears to cause rapid rearrangement of central gate residues and gate opening on the nanosecond time scale; helix hydration via water influx proceeds within 10 to 100 μs, which (in conjunction with helix 2 movement) opens an inner gate, allowing formation of the cation-conducting pore (43, 44) (Fig. 1).…”
