Tfap2a and 2b act downstream of Ptf1a to promote amacrine cell differentiation during retinogenesis

Abstract: Retinogenesis is a precisely controlled developmental process during which different types of neurons and glial cells are generated under the influence of intrinsic and extrinsic factors. Three transcription factors, Foxn4, RORβ1 and their downstream effector Ptf1a, have been shown to be indispensable intrinsic regulators for the differentiation of amacrine and horizontal cells. At present, however, it is unclear how Ptf1a specifies these two cell fates from competent retinal precursors. Here, through combined… Show more

“…Because tfap2a and tfap2b function redundantly in the development of other tissues, we next determined whether these two factors acted similarly in the pronephros (Knight et al, 2005;Van Otterloo et al, 2018;Seberg et al, 2017;Bassett et al, 2012;Jin et al, 2015). To interrogate this, we performed combination knockdown and mutant studies, and assayed expression of the solute transporters that characterize the distal nephron segments at 24 hpf.…”

“…Foxn4 acts in early-stage RPCs in conjunction with Rorb to activate expression of Ptf1a, which functions upstream of Tfap2a/b to drive amacrine cell specification (Jin et al, 2015; Liu et al, 2013). Downstream of these events, other factors act to control the specification of amacrine cell subtype.…”

“…Around E8, AP-2a is expressed in the INL, shown both in this study and by others, 33 and has been implicated in HC and AC differentiation downstream of Ptf1a. 25,34 Moreover, FoxN4 has also been suggested to regulate the generation of ACs and HCs by acting upstream of Ptf1a. [35][36][37] Strikingly, our results show that in the dsMab21l2-electroporated retina, AP-2a is severely reduced, with only a few remaining AP-2aþ cells.…”
