TGF-  and p53 Staining in CT-Guided and Endoscopic Ultrasound Fine-Needle Aspirates of Pancreatic Adenocarcinoma

Abstract: Our purpose was to determine if fine-needle aspirates of pancreatic adenocarcinoma would produce material amenable to tumor marker staining and to correlate the expression of TGF-beta and p53 with patient and tumor data. One hundred twenty specimens were analyzed. TGF-beta was positive in 26% of cases and had no correlation with patient's age, sex, survival, stage, grade, or size. p53 was positive in 22% of the cases and correlated only with grade 1 tumors. Expression of TGF-beta and p53 can be tested on prese… Show more

“…In most series, the presence of TGF-β1 is associated with shorter postoperative survival [34], although in some other studies TGF-β1 expression showed a significant correlation with longer survival time [19,29]. On the other hand, there are reports indicating that TGF-β expression has no association with survival [39]. In the present study, to have the same treatment protocol only the patients which had RO resections were considered in survival analysis and TGF-β1 and p27 expression did not show a significant influence on the survival of patients.…”

“…Only one study demonstrated the feasibility of immunocytochemistry for the assessment of p53 and transforming growth factor-b expression in the cytology samples stored as slides at the time of initial diagnosis by EUS-or CT-guided FNA. 182 The genetic profile of EUS-FNA samples has been analyzed through macro-array analysis that included 1,176 genes associated with cancer, by using commercially available complementary DNA samples. 183 A complex statistical analysis was performed (hierarchical clustering, principal component analysis, significance analysis of microarrays) that allowed initial establishment of the gene expression.…”

“…Recently we have started to focus more on the importance of cell-block preparation and now we dedicate 1 full pass for that purpose. Additional samples are sent in as needed for microbiology cultures and other ancillary studies [7]. Cell-block material was fixed in 10% buffered neutral formalin and the sections (4–5 µm) from paraffin-embedded cell block were cut and stained with hematoxylin and eosin.…”
