The ABC-Type Efflux Pump MacAB Protects Salmonella enterica serovar Typhimurium from Oxidative Stress

Abstract: Multidrug efflux pumps are integral membrane proteins known to actively excrete antibiotics. The macrolide-specific pump MacAB, the only ABC-type drug efflux pump in Salmonella, has previously been linked to virulence in mice. The molecular mechanism of this link between macAB and infection is unclear. We demonstrate that macAB plays a role in the detoxification of reactive oxygen species (ROS), compounds that salmonellae are exposed to at various stages of infection. macAB is induced upon exposure to H2O2 and… Show more

“…The macrolide-specific ABC exporter MacAB in Salmonella is induced upon exposure to H 2 O 2 and is essential for survival of S. enterica serovar Typhimurium against oxidative stress. Reactive oxygen species-mediated killing in macrophages is alleviated by MacAB (907). Both SmeIJK and MacABC of S. maltophilia also contribute to tolerance to oxidative and envelope stresses (582,584).…”

“…In this context, we also observed the possible involvement of TtgABC in the adaptation to heat and oxidative stress in P. putida KT2440, since the ECF-10 mutant as well as the ttgABCoverexpressing strain exhibited elevated cell densities in comparison to the wild type. The involvement of efflux pumps in such adaptation processes has been described in earlier studies; for example, the MacAB efflux pump plays a role in the detoxification of reactive oxygen species in S. enterica (53).…”

“…Efflux pumps of Salmonella, Campylobacter, Pseudomonas, and E. coli have been shown to facilitate the invasion of and replication within host cells and escape from host defense mechanisms (3,43). The MacAB transporter of Salmonella enterica also serves to protect the bacterium against reactive oxygen species produced by the host cells (44).…”
