The Arg72Pro Polymorphism of TP53 in Croatian Population

Abstract: Abstract. Single nucleotide polymorphisms of TP53 gene in human populations are expected to cause perturbation of p53 function, and therefore could influence risk and/or progress of disease. There are differences in inducing apoptosis, cell cycle arrest, transcriptional activity, protein interactions between variants in codon 72 (Arg72Pro), although the association of polymorphism and risk of cancer is controversial. The aim of our study was to investigate the frequency of alleles and genotypes of Arg72Pro pol… Show more

“…Distributions of p53 Arg72Pro genotypes in our healthy control group are almost identical to those reported for the Croatia, Germany, Italy, and other nearby countries, comparable in ethnicity and latitude [37][38][39][40][41]. Chi-square or Fischer exact tests did not reveal association between the CRC risk and p53 Arg72Pro polymorphism.…”

“…Our finding is similar to the findings of Tian et al [49] where there was an association between the cancer risk in Asian populations and TP53 Arg72Pro polymorphism. Zoric et al [50] also demonstrated that there was a positive association between cancer risk in ethnicity and p53 polymorphism Arg72Pro. The substantial ethnicity differences in Arg72Pro polymorphism may result from a genetic background and some influential environmental conditions such as sunlight.…”

“…It contains conformationally flexible motifs that bind transcriptional coactivator p300, involved in p53 activation through acetylation [46]. Furthermore, the PRD is important for regulating p53-mediated apoptosis [47,48]; rs1042522 (p53 p.R72P) is a common polymorphism in the PRD whose allele frequency differs in populations worldwide [49] and was recently associated with the CRC risk among specific ethnic groups according to a meta-analysis [50].…”
