The CCR4‐NOT complex component NOT1 regulates RNA‐directed DNA methylation and transcriptional silencing by facilitating Pol IV‐dependent siRNA production

Abstract: Small interfering RNAs (siRNAs) are responsible for establishing and maintaining DNA methylation through RNA-directed DNA methylation (RdDM) pathway in plants. Although the siRNA biogenesis is well known, it is relatively unclear about how the process is regulated. By a forward genetic screen in Arabidopsis thaliana, we identified a mutant defective in NOT1 and demonstrated that NOT1 is required for transcriptional silencing at RdDM target genomic loci. We demonstrated that NOT1 is required for Pol IV-dependen… Show more

“…Additional transcription factor genes, such as homologues to Arabidopsis WRI2, LEC2, EFM, NAC56, are also upregulated in all three reproductive tissue types, but their functions in moss are unknown and not easy to extrapolate from their functions in Arabidopsis. A homologue to CCR4-NOT complex component NOT1, which in Arabidopsis regulates RNA-directed methylation and transcriptional silencing (Zhou et al, 2020), is also elevated in the reproductive organs and the green sporophytes. AtNOT1 is necessary for example proper male germ cell development, pollen germination and embryogenesis (Motomura et al, 2020;Pereira et al, 2020).…”

“…Our work mainly focused on the cytoplasmic role of RNA deadenylases and directly demonstrated the poly(A) tail regulation of transposons using ONT-DRS. A previous report suggested a nuclear role of the CCR4-NOT complex in Arabidopsis as one of the essential elements for RdDM 45 . This is reminiscent of what is known in Drosophila that CCR4 co-transcriptionally represses transposon in association with Piwi 30 .…”
