The Central Vasopressinergic System: Examining the Opportunities for Psychiatric Drug Development

Abstract: Arginine vasopressin (AVP) is a cyclic nonapeptide synthesized exclusively by neurosecretory cells of the central nervous system (CNS). Two functionally distinct vasopressinergic systems can be defined based on differences in the sites of action and release of AVP. The peripheral vasopressinergic system encompasses the sites of action for AVP released into peripheral circulation (e.g. vascular smooth muscle, liver, kidney) from nerve terminals in the posterior pituitary. Peripherally circulating AVP is respons… Show more

“…With respect to anxiety-related behavior, opposite effects of AVP (anxiogenic) (5,29,34) and OXT (anxiolytic) (35)(36)(37) have been shown, a phenomenon that was used to confirm peptide-specific behav- The locomotor activity was not altered by any treatment, confirming the specificity of the manipulations on anxietyrelated behavior (data not shown).…”

“…In HAB rats, the naturally suppressed transcription of the AVP gene is disinhibited owing to a single nucleotide polymorphism in the promoter of this gene [93]. As AVP is anxiogenic independent of sex or physiological status [47,74,[94][95][96], the upregulated expression and release of brain AVP in HAB rats is, consequently, the basis of their hyperanxiety, as has been shown in males [93,96,97] and in lactating females [74]. It is not surprising that the upregulated brain AVP activity is also part of the underlying neuroendocrinological mechanism for the high levels of maternal behaviour in HAB mothers [58].…”

“…However, most of these central actions of AVP are mediated by the so-called pituitary AVP receptor, referred to as V1b or V3R. In general, V2R is much less abundant in the brain than V1a and V1b, and very little is known regarding the role of V2R in the brain (98). Little data exist on the possible function of V2R in the CNS.…”
