The Chaperonin Containing TCP1 Complex (CCT/TRiC) Is Involved in Mediating Sperm-Oocyte Interaction

Abstract: Background: Mammalian fertilization is initiated by sperm-zona pellucida (ZP) interactions. Results: The chaperonin-containing TCP1 complex (CCT/TRiC) was identified on the surface of mouse spermatozoa and shown to have an indirect role in ZP adhesion.

“…While the confirmation and significance of such targets remain to be validated, many of these substrates may be homologous to those that are activated during the capacitation of mammalian spermatozoa [33]. For instance, molecular chaperones are targeted for tyrosine phosphorylation during the latter stages of capacitation and subsequently play a key role in surface remodelling events that prime spermatozoa for their interaction with the outer vestments of the ovum [20,25,34]. Conversely, both HSPA5 and HSP90AA1 are among a cohort of molecular chaperones that reside on the human sperm surface [35] and form part of the sensing machinery that modulates intracellular calcium concentrations [36], and hence facilitates sperm capacitation.…”

“…In situ proximity ligation assays (PLA) were conducted according to manufacturer's instructions (OLINK Biosciences, Uppsala, Sweden; as described by Dun et al [25]) using anti-ODF2 and anti-phosphotyrosine antibodies. Coverslips were mounted as previously described for immunocytochemistry and visualized by fluorescence microscopy (Carl Zeiss, Sydney, Australia).…”

“…Seminal plasma transferrin correlates with sperm counts and fertility in men (Foresta et al 1986) and lactotransferrin is associated with sperm motility in Asian elephants (Kiso et al 2013). TCP1-containing chaperonins (CCTs) are present on the sperm surface of mice and boars (Belleannée et al 2011) and mediate sperm-oocyte interaction (Dun et al 2011). Interestingly, the presence of CCTs 5 and 8 in bull sperm membrane was negatively correlated with fertility.…”

“…Genes upregulated are significantly enriched for targets displaying testis-specific expression (Figures 1 and 2a; P = 0.001, Fisher's exact test; odds ratio = 1.48; 95% confidence interval = 1.16-1.86), whereas the set of genes downregulated after mating displays a deficit of testisspecific genes (P = 2.2e − 16, Fisher's exact test; odds ratio = 0.16; 95% confidence interval = 0.08-0.28). The set of upregulated genes also includes a strong enrichment for Chaperonin-containing T-complex (GO:0005832; 7 genes, P = 3.40e − 7), a complex that is known to be relevant for spermatogenesis (Soues et al, 2003;Dun et al, 2011). For instance, this category includes the genes T-cp1zeta, T-complex chaperonin 5, sarah and CCT-γ that are involved in meiosis and unfolded protein response.…”
