The Chromosomal Passenger Complex Is Required for Chromatin-Induced Microtubule Stabilization and Spindle Assembly

Abstract: In cells lacking centrosomes, such as those found in female meiosis, chromosomes must nucleate and stabilize microtubules in order to form a bipolar spindle. Here we report the identification of Dasra A and Dasra B, two new components of the vertebrate chromosomal passenger complex containing Incenp, Survivin, and the kinase Aurora B, and demonstrate that this complex is required for chromatin-induced microtubule stabilization and spindle formation. The failure of microtubule stabilization caused by depletion … Show more

“…The CPPs Survivin, Aurora B, INCENP and Borealin sequentially colocalize to the chromosome centromeres, the spindle midzone, the cleavage furrow and finally to the midbody as mitosis progresses (Adams et al, 2001a, b;Gassman et al, 2004;Sampath et al, 2004). The CPPs are already expressed at the G2/M transition and, at least in the case of Survivin, expression at earlier cell cycle stages has been reported in different types of both normal and tumor cells (Krysan et al, 2004;Song et al, 2005).…”

“…Like INCENP, Survivin has been reported to constitute both a substrate and an activator for Aurora B (Chen et al, 2003;Wheatley et al, 2004). Borealin, which was independently isolated as DasraB, is the most recently identified and the least characterized subunit of the chromosomal passenger complex (Gassman et al, 2004;Sampath et al, 2004). Borealin is phosphorylated by Aurora B in vitro but, unlike Survivin and INCENP, does not appear to increase the activity of the kinase (Gassman et al, 2004).…”

“…These are molecules of the chromosomal passenger complex (CDCA8 and Aurora kinase B; ref. 34 37,38), and several members of the kinesin superfamily of mitotic motor proteins involved in spindle function and cytokinesis (KIFC1, KIF23, KIF11, and KNSL7; refs. 39 -45).…”

“…We therefore asked whether the effects seen on this pathway upon depletion of Sgo could be specifically linked to any of these two kinases. Aurora B is present in Xenopus extracts in the so-called chromosomal passenger complex (CPC), together with INCENP, survivin, and Dasra/ Borealin (Sampath et al 2004). The CPC is required for proper localization of Sgo and Bub1 in early mitosis (Vigneron et al 2004;Resnick et al 2006;Boyarchuk et al 2007).…”
