The combination of morphogenic regulators BABY BOOM and GRF-GIF improves maize transformation efficiency

Abstract: Transformation is an indispensable tool for plant genetics and functional genomic studies. Although stable transformation no longer represents a major technology bottleneck in maize, there is still need for easily accessible and efficient transformation methods in most academic labs. Here we present the GGB transformation system, a rapid and highly efficient transformation system optimized for the immature embryo transformation of two maize genetic backgrounds, including the inbred line B104. The combination o… Show more

“…However, currently generating genome edits in monocots is challenging, time consuming and expensive. Fortunately, improvements to transgenesis are constantly improving making achieving these goals more likely in the future (43). It’s also important to consider that mutational analysis of CREs is not straightforward, often requiring numerous editing events of the cis -regulatory landscape of each gene.…”

“…This substitution may result in higher levels of GRF4‐GIF1 expression and consequently further enhance the efficiency of sorghum transformation process. A recent study proved that the combination of GRF‐GIF‐BBM could further improve maize transformation efficiency (Chen et al., 2022). We speculate that this strategy may also be effective for sorghum.…”

“…The GRF4-GIF1 are fused with CRISPR/Cas9 to produce edited plants with higher regeneration efficiency (Debernardi et al 2020 ). Recently a GGB transformation system was established with sevenfold higher transformation efficiency in maize (Chen et al 2022 ). In the GGB transformation system, the BABY BOOM transcriptional regulator (ZmBBM/EREB53) and the wheat GRF4-GIF1 (GROWTH REGULATING FACTOR4-GRF-INTERACTING FACTOR1) are combined to boost the regeneration efficiency without any side effects on plant development.…”
