The complete nucleotide sequence of RNA 1 of a German isolate of barley yellow mosaic virus and its comparison with a Japanese isolate

Abstract: The nucleotide sequence of RNA 1 of a German isolate of barley yellow mosaic virus has been determined and compared with a Japanese isolate of the same virus. The sequence identity is 93.6% at the nucleotide level and 96 % at the amino acid level. Similar values have been found for the polyproteins of the RNA 2 of both isolates (95 %). Both isolates show an RNA 1-encoded protein arrangement similar to that of potyviruses such as tobacco etch virus. In contrast, the polyproteins of the small RNAs (RNA 2) do not… Show more

“…The numerous and economically important aphid-transmitted viruses belong to the genus Potyvirus (the type species is potato virus Y) and are the most thoroughly characterized viruses of the family due to the numerous virus species for which complete sequences are available. During the past decade, it has become apparent that fungus-transmitted, bipartite viruses, such as barley yellow mosaic virus (BaYMV), share an evolutionary relationship with the aphid-transmitted potyviruses (20,23) and are members of the genus Bymovirus (the type species is BaYMV) within the family Potyviridae. The whitefly-transmitted sweet potato mild mottle virus (SPMMV) is clearly a member of the family Potyviridae, and the genus "Ipomovirus" has been proposed, with SPMMV as the sole species proposed (12).…”

“…However, studies on bymoviruses have so far been focussed on the sequence analysis of the viral genomes and electron microscopy of infected plants (Huth et al, 1984;Kashiwazaki et al, 1990;Davidson et al, 1991;Peerenboom et al, 1992;Schenk et al, 1993), and nothing is known about the nucleic acid-binding characteristics of any bymoviral CP.…”

“…Barley yellow mosaic virus (BaYMV) and barley mild mosaic virus (BaMMV) are two members of this genus that cause, often in mixed infection, the economically important yellow mosaic disease of winter barley (Hordeum vulgare) in Europe and Asia Huth & Adams, 1990 ;Hariri et al, 1990). Bymovirus research initially focused on the type species, BaYMV, whose entire genomic sequence was published for Japanese and German isolates (Kashiwazaki et al, 1990(Kashiwazaki et al, , 1991Davidson et al, 1991 ;Peerenboom et al, 1992). Since then, progress on the study of its molecular biology has been hampered by the lack of infectious clones to enable reverse genetics.…”
