Attention deficit hyperactivity disorder (ADHD) is a common neuro-developmental disorder. The symptoms of ADHD include difficulty in attention, memory and impulse control. Many pharmaceutical formulations (stimulants and non-stimulants) are available on the market to treat ADHD symptoms. The most commonly used drugs for treatment are amphetamine, methylphenidate, atomoxetine, bupropion, guanfacine and clonidine. In the field of pharmaceuticals, bioanalysis is an important tool used for the quantification of drugs and their metabolites present in biological samples using various analytical methods. Although a number of analytical methods were reported for the quantification of these drugs in biological samples of experimental animals, due to species differences, it is important to develop analytical methods to quantify these drugs in human biological samples to aid forensic and pharmacokinetic studies. In this review, we compile the bio-analytical methods such as spectrophotometry, spectrofluorimetry, mass spectrometry, electrophoresis, liquid chromatography and gas chromatography used for the quantification of ADHD drugs in human biological samples such as blood, plasma, serum, oral fluids, sweat, hair and urine based on earlier published articles from various journals.

Biological sample pretreatment is an important step in biological sample analysis. Due to the diversity of biological matrices, the analysis of target substances in these samples presents significant challenges to sample processing. To meet these emerging demands on biopharmaceutical analysis, this paper summarizes several new techniques of on-line biological sample processing: solid phase extraction, solid phase micro-extraction, column switching, limited intake filler, molecularly imprinted solid phase extraction, tubular column, and micro-dialysis. We describe new developments, principles, and characteristics of these techniques, and the application of liquid chromatography–mass spectrometry (LC–MS) in biopharmaceutical analysis with these new techniques in on-line biological sample processing.

The growing use of antidepressants in recent years has led to their increasing presence in forensic analyses. In this work, microextraction by packed sorbent followed by ultra-performance liquid chromatography with photodiode array detection provided a fast method for determining the antidepressants mirtazapine, venlafaxine, escitalopram, fluoxetine, fluvoxamine, and sertraline in human urine. The microextraction conditions (viz., type of sorbent, number of draw-eject extraction cycles or strokes, sample volume and pH, and type and volume of washing solution and eluent) were optimized by using an experimental design. The ensuing analytical method was validated in terms of linearity (25-1000 ng/mL urine), limit of detection (lower than 7.1 ng/mL), limit of quantification (25 ng/mL), precision (4.7-15.1% as relative standard deviation), and accuracy (80.4-126.1% as mean recovery for four replicate determinations). The proposed method allowed the six target antidepressants to be determined at concentrations from therapeutic to toxic levels. The application to small volumes (300 μL) of urine afforded fast extraction of the analytes and provided results on a par with those of existing clinical and forensic alternatives.
