The Development of Electrochemical Aptasensor Based on DNA Aptamers Modified by Redox Markers for Detection of Leukemia Jurkat Cells

Abstract: Oncological diseases belong to the most serious illnesses with high mortality. The most common cancer in children is acute lymphoblastic leukemia (ALL). It is important to develop diagnostic methods that will be able to detect this disease in early stage. One of the possible options can be non-invasive diagnostics using the biosensors based on nucleic acid aptamers. Aptamer recognizes the surface markers on the membrane of cancer cells with the high binding affinity. Biosensors based on aptamers with redox mar… Show more

“…For acute lymphoblastic leukemia, an oncological disease prevalent in children, a redox-labeled electrochemical aptasensor that specifically detects the cancer biomarker protein tyrosine kinase 7 (PTK7) was developed [153]. Detection was performed using Jurkat leukemia cells that overexpress PTK7, and for this purpose, two aptasensors with different redox markers (MB or Fc carboxylic acid) were described.…”

“…This approach has been used for the detection of Jurkat cells by sgc8c aptamers modified at the 3 end by ferrocene (Fc) or methylene blue (MB). The aptamers were immobilized on the gold surface by chemisorption or biotinneutravidin methods [103,106]. The redox potential for these probes is different.…”

“…A nanostructure is a form of electrode that has the potential to become a biosensor due to its high sensitivity and accuracy in detecting cancer biomarkers and its nature attractiveness with molecules [19]. Gold (Au) is commonly used as a biosensor [20][21][22]. However, Ag is also a good candidate for biosensors.…”
