Tumor Necrosis Factor-alpha (TNF-α) is one type of pro inflammatory cytokines produced by macrophages and T cell as response to acute inflammation and trauma. Exercise can be the cause of the physical stress that can lead to muscle and joints damage. Damaged muscles and joints which caused by exercising will be responded by the body inflammatory system to release pro inflammatory cytokines. Zumba fitness is an exercise that combines dance and aerobic steps. It is an application form of High Intensity Interval Training. The objective of this research is to determine the TNF-αprofile of medical students of Sam Ratulangi University shortly after zumba exercise. This research is an experimental study using post test design. The respondents are 18 medical students of Sam Ratulangi University batch 2014 who met the inclusion criterias. Blood samples were taken as soon as the respondents finished 1 hour of zumba exercise. TNF-α levels were measured with an enzyme-linked immunosorbent essay (ELISA) Quatikine ® Human TNF-𝛼. Based on the research the highest level of TNF-α is 97,59 mg/mL and the lowest level is 34,25 pg/mL with average levels of TNF-α is 75,27 pg/mL. It can be concluded that the levels of TNF-α of all subjects lay on normal range (10 pg/mL – 100 pg/mL).Keywords: TNF-α, immune system, zumbaAbstrak: Tumor Necrosis Factor-alpha (TNF-α) merupakan salah satu jenis sitokin pro-inflamasi yang diproduksi oleh makrofag dan sel T sebagai respon inflamasi akut atau respon terhadap trauma. Olahraga dapat menjadi penyebab stres fisik yang dapat mengakibatkan cedera otot maupun sendi. Kerusakan otot dan sendi yang diakibatkan karena berolahraga akan direspon oleh tubuh dengan dirangsangnya sistem inflamasi sehingga dilepaskan sitokin pro-inflamasi. Senam zumba adalah salah satu jenis olahraga aerobik yang menggabungkan tarian dan unsur aerobik dan merupakan bentuk penerapan dari metode High Intensity Interval Training, yakni latihan kardio yang dilakukan dalam waktu singkat dengan intensitas yang tinggi.Tujuan penelitian ini yaitu untuk mengetahui profilTNF-αpada mahasiswa Fakultas Kedokteran UNSRAT sesaat setelah melakukan senam zumba. Penelitian ini bersifat eksperimental lapangan dengan menggunakan rancangan post test design. Subjek penelitian berjumlah 18 orang yang telah memenuhi kriteria inklusi.Sampel darah diambil segera setelah senam zumba selama 1 jam. Kadar TNF-𝛼 diukur dengan menggunakan enzyme-linked immunosorbent essay (ELISA) Quatikine ® Human TNF-𝛼.Hasil penelitian ini menunjukkan kadar TNF-αtertinggi yaitu 97,59 mg/mL dan kadar TNF-𝛼terendah yaitu 34,25 pg/mL.
