The effectiveness of eugenol against cisplatin-induced ototoxicity

Abstract: The study findings represent the first indication in the literature that eugenol may protect against ototoxicity by raising levels of antioxidant enzymes and lowering those of oxidant parameters.

“…Eugenol suppresses the production of superoxide anion by neutrophils via the inhibition of the Raf/MEK/ERK1/2/p47phox-phosphorylation pathway [62]. Moreover, it protects against ototoxicity and neurotoxicity induced by cisplatin and aluminium, respectively, via enhancing the activities of antioxidant enzymes and lowering the lipid peroxidation and 8-OHdG levels [63,64]. In vitro, the pretreatment of HCT116 cells with eugenol, 2 h prior to the mycotoxin citrinin exposure, significantly inhibited ROS generation, modulated the antioxidants enzymes activities, and reduced MDA production, protein-bound sulfhydryls level, and DNA fragmentation [65].…”

“…They were fed with standard laboratory food and water ad libitium . Eugenol and cisplatin doses were selected from doses used by Kilic et al (), Magalhaes et al (), Sakat et al (). In our study, 24 male albino Sprague Dawley albino rats were divided into four main groups ( n = 6): In the control group, no treatment was applied. Single dose of cisplatin (15 mg/kg b.wt) was administered intraperitoneally to cisplatin group. Eugenol (100 mg/kg b.wt./day) was given to eugenol group intraperitoneally daily for 5 days. Eugenol (100 mg/kg i.p.…”

“…Histopathological samples were stained with haematoxylin–eosin (HE) and assayed by light microscopy. (Leica DM 1000, Germany) The histopathological examination method used in this study was motivated in accordance with the method used in previous studies (Kilic et al, ; Sakat et al, ). Due to histopathological findings, the sections were evaluated as no (−), mild (+), moderate (++) and severe (+++).…”
