The emerging link between cancer, metabolism, and circadian rhythms

Abstract: The circadian clock is a complex cellular mechanism that, through the control of diverse metabolic and gene expression pathways, governs a large array of cyclic physiological processes. Epidemiological and clinical data reveal a connection between the disruption of circadian rhythms and cancer that is supported by recent preclinical data. In addition, the use of animal models and molecular studies indicate emerging links between cancer metabolism and the circadian clock. This has implications for therapeutic a… Show more

“…HCC is the most common malignancy of primary liver cancers. Previous studies suggest the association of disruption of circadian rhythms with cancer development in various organs including the liver . Kettner et al generated Cry1 −/− Cry2 −/− mice, Per1 −/− Per2 −/− mice, and Alb ‐Cre‐mediated liver‐specific BMAL1 knockout mice and maintained them for 90 weeks .…”

“…Previous studies suggest the association of disruption of circadian rhythms with cancer development in various organs including the liver. 77 Kettner et al generated Cry1 −/− Cry2 −/− mice, Per1 −/− Per2 −/− mice, and Alb-Cre-mediated liverspecific BMAL1 knockout mice and maintained them for 90 weeks. 78 These transgenic mice had lower survival rates with various disorders including neurodegeneration, cystic renal dysplasia, liver steatosis, and HCC compared to wildtype mice.…”

“…This will be of great importance for breast cancer treatment and prevention if validated. The striking results obtained by Masri et al [29] and Masri and Sassone-Corsi [30] illustrated a previously unappreciated role played by lung adenocarcinoma as an endogenous circadian organizer in the rewiring of circadian homeostasis of the liver. Another important observation regarding the tumor macroenvironment and the biological clock in cancer was made by de Assis et al [31].…”

“…The circadian clock system ensures a temporal coordination of body physiology and metabolism via behavioural, neural and endocrine pathways ( Figure 1A,B). 1,5,7,[45][46][47][48][49][50][51][52][53] Peripheral oscillators operative in liver, skeletal muscle, adipose tissue, thyroid gland, endocrine pancreas and other organs regulate physiological functions within these organs, adjusting them on a daily basis to rest-activity and feeding-fasting cycles ( Figure 1B,C; Figure 2, left panel). 48,[54][55][56][57] Circadian oscillators exert their regulatory effects at several levels, from gene transcription to organ function.…”
