The Epstein-Barr virus R transactivator (Rta) contains a complex, potent activation domain with properties different from those of VP16

Abstract: Rta, encoded by Epstein-Barr virus (EBV), is a potent activator of transcription via enhancer sequences located upstream of several viral genes. To identify the domains of Rta that facilitate transcription by interacting with cellular transcription factors, different segments of Rta were linked to the DNA binding domain of yeast transactivator GAL4 (residues 1 to 147). These GALA-Rta fusion proteins were tested in transfected cells for their ability to activate the adeno Elb promoter with an upstream GALA DNA … Show more

“…Expression of downstream genes that are more strongly enhanced by AP-1(A/S) than by ZEBRA could be influenced by the higher levels of the Rta transcription factor acting in synergy with AP-1(A/S) (32). Rta is known to have an extremely potent transcriptional activation domain (43). However, Rta itself is not responsible for activation of expression of EBV late genes (Fig.…”

“…BRLF1 is translated from a bicistronic mRNA which also encodes another IE gene product, BZLF1 (12,20). These two EBV gene products function in activating expression from a number of viral (3,5,8,13,15,18,28) and perhaps cellular (7) promoters, thereby initiating the lytic cascade. However, HVS R RNA is expressed as an early transcript in HVS-infected cells (23,25).…”

“…Activation by Zta in the absence of DNA binding has also been observed with certain targets (28). The Rta transcriptional activator encoded by BRLF1 (38) also binds DNA as a dimer (34,54) and contains a powerful negatively charged, VP16-like activation domain (39). Rta can contact both TBP and TFIIB (53).…”
