The evolutionary and integrative roles of transthyretin in thyroid hormone homeostasis

Abstract: In larger mammals, thyroid hormone-binding plasma proteins are albumin, transthyretin (TTR) and thyroxine (T4)-binding globulin. They differ characteristically in affinities and release rates for T4 and triiodothyronine (T3). Together, they form a 'buffering' system counteracting thyroid hormone permeation from aqueous to lipid phases. Evolution led to important differences in the expression pattern of these three proteins in tissues. In adult liver, TTR is only made in eutherians and herbivorous marsupials. D… Show more

“…The intra-subject variability in TSH concentrations suggests another consequence of the aging process: the increased physiological heterogeneity of elderly subjects (57). The significant decrease in the TTR concentration observed in the elderly supported the idea that alterations of thyroid status occur with aging in that TTR is a plasmatic protein that binds and carries the thyroid hormones in the circulating system (58). The increased TSH concentration was probably a sign of subtle dysfunction in the regulation of the thyroid hormone production occurring with advancing age.…”

“…In addition, we have identified C40H1.5, which encodes a transthyretin-like molecule (Eneqvist et al, 2003). Studies in the mammalian system suggest that transthyretins are involved in transport of thyroid hormones (Schreiber, 2002). Also identified were inx-8 (ZK792.2) and inx-9 (ZK792.3), which encode invertebrate gap junction molecules.…”

“…The concentration of HSA is about 600 mM in the normal human serum (Schreiber, 2002). The K d values of PFOS and PFOA with the HSA are 45 mM and 3.7 mM, respectively (Chen and Guo, 2009).…”
