Background Tongue squamous cell carcinoma (TSCC) is characterized by aggressive invasion and poor prognosis. Currently, immune checkpoint inhibitors may prolong overall survival compared with conventional treatments. However, PD1/PDL1 remain inapplicable in predicting the prognosis of TSCC; thus, it is urgent to explore the genetic characteristics of TSCC. Materials and methods We utilized single‐sample gene set enrichment analysis (ssGSEA) to classify TSCC patients from the TCGA database into clusters with different immune cell infiltrations. ESTIMATE (immune‐related scores) and CIBERSORT (immune cell distribution) analyses were used to evaluate the immune landscape among clusters. GO, KEGG, and GSEA analyses were performed to analyze the different underlying molecular mechanisms in the clusters. Based on the immune characteristics, we applied the LASSO Cox regression to select hub genes and construct a prognostic risk model. Finally, we established an interactive network among these hub genes by using Cytoscape, and a pan‐cancer analysis to further verify and decipher the innate function of these genes. Results Using ssGSEA, we constructed three functional clusters with different overall survival and immune‐cell infiltration. ESTIMATE and CIBERSORT analyses revealed the different distributions of immune cells (T cells, B cells, and macrophages) with diverse immune‐related scores (ESTIMATE, immune, stromal, and tumor purity scores). Moreover, pathways including those of the interferon‐gamma response, hypoxia, and glycolysis of the different subtypes were investigated to elucidate their involvement in mediating the heterogeneous immune characteristics. Subsequently, after LASSO Cox regression, a signature of 15 immune‐related genes was established that is more prognostically effective than the TNM stage. Furthermore, three hub genes—PGK1, GPI, and RPE—were selected using Cytoscape evaluation and verified by immunohistochemistry. PGK1, the foremost regulator, was a comprehensively profiled pan‐cancer, and a PGK1‐based interactive network was established. Conclusion Our results suggest that immune‐related genes and clusters in TSCC have the potential to guide individualized treatments.

Introduction: Cutaneous squamous cell carcinoma (cSCC) is one of the most frequently occurring types of cancer in humans. Scientometric research is an innovative method for analyzing the research trends in various domains, with great implications in the field of medicine. Materials and Methods: We searched the Web of Science database with the following established query terms: “Squamous cell carcinoma”, “skin”, and “immunohistochemistry”. After applying the inclusion and exclusion criteria, a total of 76 articles were selected. The present study aims to analyze, based on the frequency of use of keywords with scientometric algorithms and map-based distributions, the trends of the research concerning cSCCs in 2017–2022. Results: A graphical representation based on 11 scientometric maps presented the division of the keywords into seven clusters, from which seven categories of research interest were defined. The clusters represent a multidisciplinary approach to the diagnosis and treatment of cSCCs, cancer diagnostics, patient outcomes, histopathological importance, management of cSCCs, role of progression, and adequate treatment of and importance of immunohistochemistry for cSCCs. The distribution of the citations shows the importance of the available research on cSCCs by analyzing the first five most-cited articles included in our study in direct concordance with the seven defined clusters. Conclusion: The scientometric research method reveals the interest of research in the multidisciplinary approach used to obtain the best outcomes for the patient, including a targeted investigation, as well as diagnostic and treatment options. The trends in the research reveal that histopathological diagnostics and immunohistochemistry, combined with molecular techniques, are the most important tools used to establish a personalized diagnosis, thus increasing the quality of life and life expectancy for patients with cSCCs.

The article presents data concerning effectiveness of the immune agent alpha/betadefensins as a accompanying chemotherapy/radiotherapy drug at the 1st stage oftreatment of 73 patients with cancer of the oral cavity and oropharynx.The purpose of the work – to evaluate the effectiveness of the immune agent alpha/betadefensins as a drug, accompanying radiation or chemoradiation therapy, at the 1st stageof treatment of patients with the oral cavity and oropharynx cancer.Materials and methods. Patients of the I and II groups under study received radiation andchemoradiation therapy, respectively, as well as the immune agent alpha/beta defensins2.0 ml 2 times a day intramuscularly 2 days before the start of the special treatment for5 days, as well as during the next 10 days of treatment 1 time per day. Patients of III-IVgroups, comparative, received similar treatment, but without immunotherapy.The results. According to the time of the appearance of epitheliitis and changes in itsdegree in the dynamics of treatment, the best results were obtained in patients of groupI, since the first inflammation manifestations in the oral cavity or oropharynx developedat the latest – at a dose of 22 Gy, while in patients of group III they appeared at a dose of12 Gy (p<0.05) and in larger quantity of patients (12%). The transition of the first degreeof epitheliitis to II and III in the first group took place in a smaller number of patients,that indicates a positive effect of the immunopreparation on the severity of epitheliitis.The number of cases of epitheliitis of the III degree is indicative: 4% of patients of theI group against 79% of patients of the III group and 60% of patients of the IV on averageafter 30 Gy (p<0.05). Only in the IV group there were patients who developed ulcerativeepitheliitis (IV degree). The number of patients with radiodermatitis of the I degree in theI group was 30% less compared to the III group, in the II group – 29% less than in groupIV. Radiodermatitis of the II degree was observed in a small number of patients – 5% inthe II group and 11% in the III group Xerostomia occurred in 90% of patients regardlessof the method of treatment and the use of immunotherapy as accompanying drug ofspecial treatment. Paraheusia and ageusia are more pronounced in groups III and IV.Conclusions. The phenomena of radiation epitheliitis develop later in the treatmentperiod and in a smaller number of patients, who received alpha/beta defensinimmunotherapy compared to patients, who did not receive immunotherapy. A positiveeffect of immunotherapy can also be considered a decrease in the number of patients withIII-IV degrees of epitheliitis.
