The expression of the ubiquitin ligase subunit Cks1 in human breast cancer

Abstract: Introduction Loss of the cell-cycle inhibitory protein p27 Kip1 is associated with a poor prognosis in breast cancer. The decrease in the levels of this protein is the result of increased proteasome-dependent degradation, mediated and rate-limited by its specific ubiquitin ligase subunits S-phase kinase protein 2 (Skp2) and cyclin-dependent kinase subunit 1 (Cks1). Skp2 was recently found to be overexpressed in breast cancers, but the role of Cks1 in these cancers is unknown. The present study was undertaken t… Show more

“…37 Additionally, in vitro models have identified Cks1 as an essential cofactor for efficient Skp2-dependent ubiquitination of p27 Kip126 and its overexpression was found strongly associated with poor prognosis in carcinomas of the colorectum, stomach, and breast. 28,29,38 Furthermore, gene amplification and overexpression of Cks1 at chromosome band 1q21 is associated with reduced levels of p27 Kip1 and an aggressive clinical course in multiple myeloma. 39 In our univariate analysis, Cks1 overexpression also correlated with inferior OS in gallbladder carcinomas, although it did not remain prognostically independent in multivariate comparison.…”

“…Increases in Cks1 expression have been shown in a number of human malignancies (1)(2)(3)(4). Increase in Cks1 mRNA has also been shown in PyMT-initiated mammary tumors (17), and in urethane and ENU-initiated lung tumors in rasH2 mice (32).…”

“…Kip1 (1). Interestingly, high expression of Cks1 has also been reported in non-small cell lung carcinomas, although it was not inversely related with p27 Kip1 expression, suggesting that Cks1 may mediate some of its functions via Skp2 and p27 Kip1 -independent mechanisms (3).…”

“…In fact, overexpression of Skp2 and/or Cks1 has been found in breast cancer tissues and is an independent prognostic marker for disease-free and overall survival and is associated with aggressiveness (5-9). In cell culture, higher expression levels of Skp2 were found in estrogen receptor (ER)/Her2-negative aggressive breast cancer cells, such as MDA-MB-231, as compared to MCF-7 ER-positive breast cancer cells (6,7).…”
