The head and neck cancer (HN-5) cell line properties extraction by AFM

Abstract: This work incorporates experimental methods based on Atomic Force Microscopy (AFM) in order to extract the physical and mechanical characteristics of the head and neck cancer (HN-5) cell line such as cell topography, modulus of elasticity and viscoelastic properties. The initial parameters to determine the mechanical properties are obtained by extracting information from cantilever's force-displacement curve and vertical and horizontal displacement. Next, the changes in elasticity modulus at different points i… Show more

“…33 for cancer diagnosis, such as head and neck cancer or ovarian cancer. 34 However, most studies only focus on the changes in the mechanical properties of single cell, but ignore the changes in that of tissues. In this study, fresh live surgical prostate specimens were collected and found that PCa tissues were softer than normal tissues, which exhibited its clinical significance.…”

“…AFM has been widely used to measure the deformability of single cell after the first detection in bladder cancer cell line since 1999 33 . Changes in the mechanical properties of single cell have been identified as one of the indicators for cancer diagnosis, such as head and neck cancer or ovarian cancer 34 . However, most studies only focus on the changes in the mechanical properties of single cell, but ignore the changes in that of tissues.…”

“…Korayem et al [ 276 ] recently used the microcantilever-based atomic force microscope to obtain various mechanical and physical properties of the head and neck cancer cells. These properties include the modulus of elasticity, cell topography, and viscoelastic properties.…”

“…Biological atomic force microscopy (Bio-AFM) is a powerful tool, particularly at microscale- and nanoscale levels ( 15 ). It is widely used in biomedical research to determine the mechanical properties of various tumor tissues including in breast cancer ( 16 ), liver cancer ( 17 ), lung cancer ( 18 ), bladder cancer ( 19 ), colon cancer ( 20 ), head and neck cancer ( 21 ), and prostate cancer ( 22 ). It is used to explore the intrinsic relationship between tumor cell proliferation, adhesion, metastasis, and the mechanical properties of tumor cells.…”
