The IgCAM CLMP regulates expression of Connexin43 and Connexin45 in intestinal and ureteral smooth muscle contraction in mice

Abstract: CAR-like membrane protein (CLMP), an immunoglobulin cell adhesion molecule (IgCAM), has been implicated in congenital short-bowel syndrome in humans, a condition with high mortality for which there is currently no cure. We therefore studied the function of CLMP in a Clmp-deficient mouse model. Although we found that the levels of mRNAs encoding Connexin43 or Connexin45 were not or were only marginally affected, respectively, by Clmp deficiency, the absence of CLMP caused a severe reduction of both proteins in … Show more

“…Connexin 43 was observed by punctate staining in the hydrogel ( Figure 2B), which is consistent with connexin 43 staining in a 2D culture control ( Figure 2C) and typical for connexin 43 (negative control images included in Figure S2, Supporting Information). [61,62] Connexin 43 comprises hemichannels and gap junctions along the cell membrane and is essential for osteocyte function, survival, and differentiation. [63][64][65] Concomitant, normalized expression (NE) of osteocyte-related genes Dmp1 and Sost, as measured by qPCR, increased over culture time ( Figure 2D).…”

“…However, the intestinal length is not reduced in mutant mice in comparison to wild-type mice as shown for the human CLMP gene. 34 Recently, Gonnaud et al described a CSBS patient with intestinal malrotation and short intestine with 35 cm length and also chronic intestinal pseudo-obstruction syndrome. They identified two previously unreported mutations in the CLMP gene.…”
