The Intergenic Region between the Divergently Transcribed niiA and niaD Genes of Aspergillus nidulans Contains Multiple NirA Binding Sites Which Act Bidirectionally

Abstract: The niaD and niiA genes of Aspergillus nidulans, which code, respectively, for nitrate and nitrite reductases, are divergently transcribed, and their ATGs are separated by 1,200 bp. The genes are under the control of the positively acting NirA transcription factor, which mediates nitrate induction. The DNA binding domain of NirA was expressed as a fusion protein with the glutathione S-transferase of Schistosoma japonicum. Gel shift and footprint experiments have shown that in the intergenic region there are fo… Show more

“…Growth on nitrate medium induces the specific niiA/niiD transcription factor and thus sasA expression, whereas growth on the universal repressor ammonium reduces activity of the general nitrogen source transcription factor areA and therefore reduces sasA expression (Arst and Cove, 1973;Punt et al, 1995). Both strains were grown on inducing nitrate medium (NO À 3 ), inducing and repressing proline, nitrate and ammonium medium (proline + NH þ 4 + NO À 3 ), repressing proline and ammonium medium (proline + NH þ 4 ) and noninducing and nonrepressing proline medium.…”

“…Nitrate and ammonium ions are known to influence nitrogen metabolism. 24) Accordingly, production of a violet pigment, PP-V, was strongly suggested to be associated with the nitrogen metabolism.…”

“…In Aspergillus nidulans, the nitrate reductase and nitrite reductase were designated niaD and niiA respectively. 25) So the nitric oxide reductase gene of A. oryzae was designated nicA. On the other hand, the deduced amino acid sequence of the gene was 58.9% identical to that of CYP55A1.…”
