Phenotypic differences among populations within a species have been reported for a variety of traits, ranging from life history to physiology to gene transcription. Population‐level phenotypic variation has been attributed to genetic differences resulting from genetic drift and/or local adaptation as well as environmental differences resulting from plasticity. We studied population‐ and family‐level variation in gene transcription for 22 fitness‐related genes, comprising immune, growth, metabolic, and stress processes in Chinook salmon (Oncorhynchus tshawytscha). We created hybrid Chinook salmon families from eight populations and treated them with an immune stimulus, a handling stress challenge, and held some as a no‐treatment control group. Population effects, sire effects, and narrow‐sense heritability (h2) were calculated for each candidate gene within each treatment group. We expected population to have a significant effect on gene transcription for many of our genes; however, we found a population effect for transcription at only one immune gene at rest. The limited number of significant population effects on gene transcription, combined with significant additive genetic variance within each population does not support the expectation of past strong selection pressures acting on heritable transcription profiles among populations. Instead, our results indicate that Chinook salmon likely adapt to their local environment through transcriptional plasticity rather than fixed differences. The expectation for fixed population‐level differences in gene transcription at fitness‐related genes, reflecting accepted models of local adaptation is high; however, comparisons among multiple populations using half‐sibling breeding designs are rare. Our work fills an important gap in our growing understanding of the process of among and within‐population divergence.

Mitochondrial DNA was isolated from the Korean freshwater gobioid fish Odontobutis platycephala by long-polymerase chain reaction with conserved primers and this mtDNA was sequenced by primer walking using flanking sequences as sequencing primers. The resultant O. platycephala mtDNA sequence was found to be 17 588 bp in size with a mostly conserved structural organization when compared with that of other teleost fish. Rearrangements of tRNAs (tRNA-Ser, tRNA-Leu, tRNA-His) and an additional non-coding region (533 bp) were present between the ND4 and ND5 genes. In the present paper, the basic characteristics of the O. platycephala mitochondrial genome is reported, including its structural organization, base composition of rRNAs, tRNAs and protein-encoding genes, characteristics of mitochondrial tRNAs and the peculiar rearrangement features of some parts of the mtDNA. Phylogenetic analysis performed using the cytochrome b gene sequences of 16 Korean freshwater fishes (15 gobioids) with the Bayesian algorithm showed that O. platycephala forms a clade (1Á00 of posterior probability) with other species of Odontobutis. This suggests that the observed rearrangement between the ND4 and ND5 genes in the O. platycephala mitogenome reflects independent events.

Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells. In vertebrates, three main groups of actin isoforms namely alpha, beta, and gamma have been identified. The alpha actins are found in muscles with contractile functions whereas beta and gamma actins co-exist in the cytoskeleton. For establishing evolutionary inter-relationship, sequence analysis and alignment of protein sequences, seems to be crucial. In the present study, a bioinformatics approach has been adopted to explore the sequence, structure and phylogenetic analysis of β-actin protein from eight commercially important fishes and compared with the standard sequences like 1HLU Bos taurus, 2OAN Bos taurus and 2BTF Bos taurus, which were predicted to be the closest homologs for the 11sequences of the β actin protein of 8 fishes. From the comparative analysis it was concluded that 1HLU Bos taurus affords highest sequence coverage with low E-value with the 11 sequences of eight fishes under study. Presence of conserved residues in the sequences and the phylogenetic relationship among fishes with respect to the β-actin protein, have been discussed.
