The isolated perfused rat liver: standardization of a time-honoured model

Abstract: SummaryFor many years, the isolated perfused rat liver (IPRL) model has been used to investigate the physiology and pathophysiology of the rat liver. This in vitro model provides the opportunity to assess cellular injury and liver function in an isolated setting. This review offers an update of recent developments regarding the IPRL set-up as well as the viability parameters that are used, with regards to liver preservation and ischaemia and reperfusion mechanisms.A review of the literature was performed into … Show more

“…These include the release of a host of potential biomarkers, including liver enzymes (AST, ALT, LDH and GGT) into the perfusate. However, these enzymes can be released from the tissue by simple manual manipulation (12,13). Consequently, their utility in evaluating the hepatotoxicity of drugs and drug candidates ex vivo is questionable.…”

“…The liver perfusion apparatus used in our laboratory is based on the description of Miller et al but whit changes [9][10][11]. The perfusion apparatus consisted of a perfused reservoir maintained at 37 °C, that perfusion was carried out by a peristaltic pump at a constant flow rate of 20 ml/min for 120 min.…”
