The Lectin LecA Sensitizes the Human Stretch-Activated Channel TREK-1 but Not Piezo1 and Binds Selectively to Cardiac Non-myocytes

Abstract: The healthy heart adapts continuously to a complex set of dynamically changing mechanical conditions. The mechanical environment is altered by, and contributes to, multiple cardiac diseases. Mechanical stimuli are detected and transduced by cellular mechano-sensors, including stretch-activated ion channels (SAC). The precise role of SAC in the heart is unclear, in part because there are few SAC-specific pharmacological modulators. That said, most SAC can be activated by inducers of membrane curvature. The lect… Show more

“…5 b). Inward bending of Gb3 + GUVs probably indicates tight interactions of Shiga-CAR T cells with Gb3-functionalised vesicles, related to membrane invaginations induced by StxB [ 72 , 73 ] or LecA in Gb3 + GUVs and HEK cells [ 44 , 74 ]. More recently, a molecular dynamics simulation also proposed the induction of negative membrane curvature triggered by StxB and LecA after binding to and clustering Gb3 [ 75 ].…”

“…Inhibiting LecA by galactose or galactose-derived compounds proved to be efficient against lung infection in murine model systems ( Chemani et al, 2009 ; Boukerb et al, 2014 ). Furthermore, LecA selectively bound to cardiac non-myocytes and altered plasma membrane topology ( Darkow et al, 2020 ).…”

“…To record ion channel activity in the plasma membrane, the patch-clamp technique in voltage-clamp mode was used as described previously [60]. Recordings were obtained in whole-cell (electrical stimulation) or inside-out (mechanical stimulation) configurations.…”

“…For overexpression of human TREK-1 plasmid, jetPEI reagent (101-10N, Polyplus transfection, France) was used according to the manufacturer's instructions and as described previously [60]. The TREK-1-internal ribosome entry site 2 (IRES2)-enhanced green fluorescent protein (eGFP) plasmid was kindly provided by Eric Honoré and Delphine Bichet.…”
