The long noncoding RNA SPRY4-IT1 increases the proliferation of human breast cancer cells by upregulating ZNF703 expression

Abstract: BackgroundLong noncoding RNAs (lncRNAs) have emerged recently as a new class of genes that regulate cellular processes, such as cell growth and apoptosis. The SPRY4 intronic transcript 1 (SPRY4-IT1) is a 708-bp lncRNA on chromosome 5 with a potential functional role in tumorigenesis. The clinical significance of SPRY4-IT1 and the effect of SPRY4-IT1 on cancer progression are unclear.MethodsQuantitative reverse transcriptase PCR (qRT-PCR) was performed to investigate the expression of SPRY4-IT1 in 48 breast can… Show more

“…Overexpression of SKA2 appears to promote proliferation and human breast cancer progression (34). Recently, microarray analysis to identify genes that exhibited a change in expression following SPRY4-IT1 knockdown in MDA-MB-231 cells, indicating that SKA2 may represent a notable downstream effector of SPRY4-IT1 (35). The present study revealed that SKA2 expressions were elevated in glioma tissues; a significant positive correlation was also observed between SPRY4-IT1 and SKA2 in human glioma tissues.…”

“…Shi et al 11 first reported that lncRNA SPRY4-IT1 is overexpressed in breast cancer tissues and cell lines. In addition, they observed that overexpression of SPRY4-IT1 is significantly correlated with tumor grade and tumor size.…”

“…Interestingly, the expression level of SPRY4-IT1 in estrogen receptor negative (ER−) breast cancer tissues is higher than that in ER positive (ER+) breast cancer tissues. To investigate the biological roles of SPRY4-IT1 in breast cancer, Shi et al 11 downregulated SPRY4-IT1 in MDA-MB-231 and MDA-MB-435S cells and overexpressed SPRY4-IT1 in MCF-7 cells. They found that knockdown of SPRY4-IT1 decreased the cell colony formation ability, promoted G1 arrest, and caused apoptosis in breast cancer cells.…”

“…Previously, multiple studies reported that in different types of cancer there was abnormal expression of lncRNA, indicating that the same lncRNA may serve different functions in different types of cancer. For instance, a lncRNA named SPRY4 intronic transcript 1 (SPRY4-IT1) was reported to be overexpressed in melanoma, renal cell carcinoma, esophageal squamous cell carcinoma, breast cancer, bladder cancer and glioma, and associated with poor prognosis and promotion of tumor growth (24)(25)(26)(27)(28)(29). Certain studies also demonstrated that SPRY4-IT1 expression was decreased in non-small-cell lung cancer and gastric cancer, and acted with significant antitumor function in these two types of cancer (30,31).…”
