The MyoD1 Promoted Muscle Differentiation and Generation by Activating CCND2 in Guanling Cattle

Abstract: The purpose of this study was to analyze the transcriptome of MyoD1 gene knockout MDBK cells (bovine kidney cells) using high-throughput sequencing. For the first time, CRISPR/CAS9 technology was used to construct a MyoD1 knockout in MDBK cells and transcriptome sequence analysis was used to examine MyoD1-related target gene expression. Transcriptome sequencing indicated the presence of 723 differentially expressed genes (DEGs) by comparing wild type and MyoD1 knockout MDBK cells and included 178 upregulated a… Show more

“…It is also thought to be a possible candidate gene for characteristics related to meat production in livestock [ 46 , 47 , 48 , 49 ]. In a relevant study conducted on Guanling cattle, it has been confirmed that MYOD1 is closely associated with muscle formation and differentiation [ 50 ]. Furthermore, MYOD1 regulates muscle growth and development by recruiting transcription factors such as c-Jun, Jdp2, Meis, and Runx1, thereby enhancing the expression of genes related to muscular characteristics [ 51 ].…”

“…Previous studies have shown the PDE1B gene could be regarded as a positional and functional candidate gene for carcass traits in beef cattle. Zhou Di et al [35] used high-throughput sequencing to analyze the MyoD1 gene knockout transcriptome of MDBK cells (bovine kidney cells) and found PDE1B was highly expressed in the vine leg muscle, dorsal long muscle, and shoulder, which was related to muscle formation and differentiation. Therefore, PDE1B has a certain promoting effect on fetal muscle formation and differentiation, and it has a positive effect on daughter pregnancy, and PDE1B can be further identified as a candidate gene for DPR.…”

“…Guanling cattle are primarily raised in Guanling Buyi and Miao Autonomous County, located in Guizhou Province. Consequently, these cattle are named after the region they originate from, known as Guanling cattle (Zhou et al 2022). The Guanling cattle breed is recognised for its resilient physique, sturdy limbs, and hooves, making it highly suitable for agricultural and grazing activities in mountainous regions (Liu 2015).…”
