For the development of renal diseases, oxidative stress (OS) is reasoned to be one of the risk factors. For the treatment or prevention of the renal disease, the use of antioxidants could be a hopeful therapeutic mediation as they retard or block the oxidative reaction along with the inflammatory process. Luteolin (Lut) is a plant flavonoid, a pharmacologically active component normally found in glycosylated forms in basic perilla leaf, green pepper, celery, seed, honeysuckle bloom, and chamomile blossom; it exhibits antioxidant activity. In this investigation, we explored the nephroprotective activity of Lut on bisphenol A (BPA)‐induced nephron toxicity in rats. Orally administering Lut (100 and 200 mg/kg) diminished BPA‐induced anomalies in the kidney, blood urea nitrogen, creatinine, and serum uric acid levels. Lut therapy reduced the BPA‐influenced generation of inflammatory mediators, inclusive of tumor necrosis factor alpha, interleukin 6, and interleukin 1 beta. This was coupled with significant improvement in kidney histopathologic features. Lut enhanced the nuclear factor‐like 2 (Nrf2) and heme oxygenase 1 (HO‐1) expression, which showed protection against OS induced by BPA. The current outcomes of the study showed that Lut has a strong reactive oxygen species scavenging property and potentially decreases the lipid peroxidation as well as inhibits DNA damage in renal toxicity induced by BPA. In conclusion, the potential antioxidant effect of Lut may be because of its modulatory effect on the Nrf2/antioxidant response element (ARE)/HO‐1 pathway, which means it protects the kidney from BPA‐induced oxidative injury. © 2019 IUBMB Life, 2019

Background Ifosfamide (IFO) is an alkylating agent administered against different types of malignancies. Several cases of renal injury and serum electrolytes disturbances have been reported in IFO-treated patients. Oxidative stress and mitochondrial dysfunction are suspected of being involved in the mechanism of IFO nephrotoxicity. Carnosine is a dipeptide which its antioxidant and mitochondria protecting properties have been mentioned in different experimental models. The current study aimed to evaluate the nephroprotective properties of carnosine against IFO-induced renal injury. Methods Rats were treated with IFO (50 mg/kg, i.p) alone or in combination with carnosine. Serum and urine biomarkers of renal injury in addition to kidney markers of oxidative stress were evaluated. Moreover, kidney mitochondria were isolated, and some mitochondrial indices were assessed. Results Elevated serum creatinine and BUN, hypokalemia, and hypophosphatemia, in addition, to an increase in urine glucose, protein, γ-GT, and alkaline phosphatase (ALP), were evident in IFO-treated animals. IFO also caused an increase in kidney reactive oxygen species (ROS) and lipid peroxidation (LPO). Renal GSH levels and antioxidant capacity were also depleted with IFO therapy. Mitochondrial dehydrogenase activity, GSH level, membrane potential, and ATP content were decreased while mitochondrial LPO and permeabilization were increased in IFO group. Carnosine (250 and 500 mg/kg, i.p) mitigated IFO-induced oxidative stress and mitochondrial impairment in renal tissue. Conclusion Our data suggest mitochondrial dysfunction and oxidative stress as fundamental mechanisms of renal injury induced by IFO. On the other hand, carnosine supplementation protected kidneys against IFO-induced injury through regulating mitochondrial function and mitigating oxidative stress.

Introduction: Misuse of anabolic androgenic steroids (AAS) has become a widespread health problem with subsequent multisystem adverse effects. However, among their adverse effects, renal toxicity remains one of the least discussed. Aim of work: To study the histopathological effects of Nandrolone Decanoate (ND) abuse on renal cortex and examine the possible protective role of taurine. Materials and methods: Thirty-six adult male albino rats, aging 4-6 months and weighing 200-250 gms, were used in this study. Animals were divided equally into three groups; Group I (control group): subdivided into Subgroup IA: negative control and Subgroup IB: rats received intramuscular 0.5ml sesame oil twice weekly for six weeks and Subgroup IC: received 100 mg/kg/day Taurine by gavage, for six weeks. Group II (ND Group): received intramuscular 5 mg/kg ND, dissolved in sesame oil, twice weekly for six weeks. Group III (Taurine and ND Group): given both Taurine and ND in the same dose and duration as subgroup IC and group II respectively. At the end of the experiment, venous blood samples were collected from the tail vein for biochemical assays. Kidneys were dissected out and processed for light and electron microscopic examination. Results: Examination of group II revealed marked congestion, obliteration of Bowman's space and lumina of both proximal and distal tubules. Ultrastructural distortion was evident in glomerular filtration barrier and tubular lining cells. Statistically significant biochemical changes were also observed. However, group III showed almost preserved glomerular and tubular architecture with few persistent changes. Biochemical results also revealed marked improvement. Conclusion: Marked renal cortical affection with renal functional deterioration were noted following ND. Taurine exerted marked reno-protection thus, it would be also of great help to protect AAS users against possible renal hazards in case their AAS use was inevitable.
