The neurodevelopmental spectrum of synaptic vesicle cycling disorders

Abstract: Neurodevelopmental disorders encompass diverse, dynamic and interactive childhood-onset symptoms, which can include sensory deficits, motor impairments, epilepsies, intellectual disabilities and mental health difficulties. Until recently, the cause of each individual's disorder was most often unknown. Now, genomic analysis can diagnose a specific cause in 60% of severely affected children (Gilissen et al., 2014), and the catalogue of genetic diagnoses associated with neurodevelopmental disorders has rapidly ex… Show more

“…Irrespective of seizure burden and independent from the pathogenic gene variation, the overall development was described as normal in 95% of patients and affected relatives. Behavioral issues were not reported, which is in line with previous reports [ 30 ]. Behavioral comorbidities were not assessed systematically.…”

“…A number of inherited variants and de novo mutations have been identified in human KIF1A from clinical studies. These mutations have been linked to neurodevelopmental and neurodegenerative disorders, including spastic paraplegias, encephalopathies, intellectual disability, autism, and sensory neuropathies (reviewed in Boyle et al, 2020 ; Gabrych et al, 2019 ; Guo et al, 2020 ; John et al, 2020 ; Nicita et al, 2020 ; Van Beusichem et al, 2020 ). In KIF1A-associated neurological disorder (KAND), the mutations span the entirety of the KIF1A protein sequence ( Boyle et al, 2020 ).…”

“…The orchestrated release of neuromodulators is essential for proper development and function of the nervous system. Variants in genes encoding for proteins involved in synaptic vesicular trafficking are increasingly recognized as a cause of neurodevelopmental disorders 8 . The importance of DCV trafficking in brain development is underlined by the fact that two protein members of the EARP complex, VPS‐51 and VPS‐53, have been associated with neurodevelopmental disorders 9‐12 .…”
