The optimal sterilizing compound and culture medium in Elaeocarpus grandiflorus L. in vitro shoot induction

Abstract: Elaeocarpus grandiflorus L. is an identity plant of Salatiga City. Wherein, the plant is difficult to naturally propagation. This study would obtain propagation technique of E. grandiflorus by tissue culture. It was experimentally conducted, consists of two sub-experiments. The first aimed to find the optimal sterilization agent was conducted with a completely randomized design, namely sterilizing compound. There were two compounds studied, i.e., NaOCl and Ca(OCl)2. The second sub-experiment aimed to determine… Show more

“…NaOCl and 70% alcohol were used in chemical solutions for surface sterilization as both are widely employed as disinfectants. Hypochlorite ions regenerated from hypochlorous acid (HOCl) and dissolved in water are capable of damaging bacterial plasma membranes (Rahayu et al 2019).…”

“…In tissue culture, the most prevalent issues are improper procedures and insufficient disinfectant levels. External contamination of explants from contaminated tools, equipment, and personnel in media preparation and culture is also a hazard (Abass, 2013;Rahayu et al, 2019;Emoghene et al, 2020).…”

“…Explants simply need to be surface sterilized by treatment with a disinfectant solution at acceptable concentrations for a defined amount of time; living materials should not lose their biological activity during sterilization, and only impurities should be removed; (Oyebanji et al, 2009). Sodium hypochlorite (NaOCl), calcium hypochlorite Ca (ClO) 2 , mercuric chloride (HgCl 2 ), silver nitrate (AgNO 3 ), and hydrogen peroxide (H 2 O 2 ) are some of the most commonly utilized sterilants (Rahayu et al, 2019).…”
