The p23 molecular chaperone promotes functional telomerase complexes through DNA dissociation

Abstract: Telomeres are the composite of short DNA element tandem arrays and heterotypic protein components that protect and maintain chromosomal termini. As proper telomere maintenance requires a multitude of DNA extension events, it is important to understand the factors that modulate telomerase DNA association. Here, we show that the endogenous levels of the yeast p23 molecular chaperone Sba1p are required for telomere length maintenance and that Sba1p can modulate telomerase DNA binding and extension activities in v… Show more

“…1, 4, and 7). This may also be the case for other prosurvival protein clients of Hsp90 that are dependent on p23 such as other steroid receptors and telomerase (45). Therefore, a thorough comparative study on gedunin's effects on normal versus cancer cells warrants further investigation.…”

“…Telomerase extracts were prepared using wild-type (YPH499) or est1⌬ Saccharomyces cerevisiae as described (24). In brief, yeast extract was sequentially resolved over DEAE and MonoQ resins (GE Healthcare Inc.) using TMG buffer (20 mM Tris pH 8.0, 1.1 mM MgCl2, 0.1 mM EDTA, 1.5 mM DTT, 10% glycerol, 0.1% Triton X-100, and NaOAc pH 5.0).…”

“…The ChIP experiments were performed as previously described (24). To determine the relative telomere occupancy by Est1 derivatives, est1⌬ transformants expressing wild-type Est1, Est1-41, or Est1-60 were grown to an O.D.595 ϭ 0.5 before formaldehyde crosslinking and immunoprecipitation.…”

“…Telomerase was partially purified from wild-type yeast (YPH499) by chromatography over DEAE-Sepharose fast flow resin (GE Healthcare) and Mono Q resin (GE Healthcare) as described previously (5,25) and used in telomerase DNA extension assays. Briefly, partially purified telomerase extract was incubated with 2 pmol of a 7-base 3Ј-overhang template (GTGTGTG) immobilized on streptavidin paramagnetic beads (Promega) and extension buffer (50 mM Tris, pH 8.0, 1 mM MgCl 2 , 1 mM spermidine, 1 mM DTT, 0.5% glycerol, 50 M dTTP, 10 Ci of [␣-…”
