The Peptidoglycan Recognition Proteins PGRPLA and PGRPLB Regulate Anopheles Immunity to Bacteria and Affect Infection by Plasmodium

Abstract: Peptidoglycan recognition proteins (PGRPs) form a family of immune regulators that is conserved from insects to mammals. In the malaria vector mosquito Anophelescoluzzii, the peptidoglycan receptor PGRPLC activates the immune-deficiency (Imd) pathway limiting both the microbiota load and Plasmodium infection. Here, we carried out an RNA interference screen to examine the role of all 7 Anopheles PGRPs in infections with Plasmodium berghei and P. falciparum. We show that, in addition to PGRPLC, PGRPLA and PGRPS2… Show more

“…We also noticed the signi cant induction of caudal, and reduction of pgrps, pgrp-la, pgrp-lc, pgrp-ld, and the antimicrobial peptides, defensin in dsAsteglut1-treated mosquitoes, suggesting that Asteglut1 might be involved in regulation of immune responses [25,40,56]. PGRP-LA is a receptor of the mosquito immune de ciency pathway (Imd) [41]. It helps to control the homeostasis of gut microbiota and parasite infection in An.…”

“…Five immune related genes were differentially regulated. Caudal, the negative regulator of Imd pathway was signi cantly upregulated (P = 0.0337) [40], while the peptidoglycan recognition proteins, pgrp-la, -lc,ld, and the antimicrobial peptides, defensin were signi cantly downregulated (P = 0.0111, P = 0.0378, P = 0.0022) [25,[40][41][42][43]. These results indicate that Asteglut1 might control parasite infection by regulating mosquito immune responses.…”

“…In the ensuing cytoplasmic cascade, NF-κB transcription factor Rel2 is phosphorylated by IKK2 and cleaved by DREDD to induce its nuclear translocation. IMD pathway is repressed by Caspar (Sim et al, 2014), an Ectoderm-expressed 4-containing module (Ect4) (Akhouayri et al, 2011) and PGRP-LB (Gendrin et al, 2017). We observed an induction of IMD pathway by DENV and ZIKV through PGRP-LB downregulation, and by CHIKV through Caspar downregulation and IKK2 upregulation (Figure 1c).…”
